硒对体外培养的大鼠皮层神经细胞形态学及NSE、NGF表达的影响

摘    要:目的 研究硒对体外神经细胞生长发育的直接影响及可能的作用机制。方法 采用体外新生大鼠皮层神经细胞培养法观察微量元素硒对神经细胞形态学及其表达NSE、NGF蛋白的影响。结果 ①中低浓度的硒能增加体外培养的神经细胞最长突起长度和细胞平均直径 ;②硒能促进体外培养的神经细胞表达NSE蛋白 ;③体外培养的大鼠皮层神经细胞随着培养时间的延长 ,NGF的表达减弱 ,与对照组比较 ,在培养的第 5、6、7天 ,硒能促进神经细胞NGF的表达。结论 硒可能是通过促进、增强NSE、NGF蛋白的表达 ,进而直接作用于体外培养的神经细胞 ,促进神经细胞的生长、发育与分化

Abstract:Objective To study and observe the effects of selenium(Se) on the growth, development,and differentiation in cultured neurons in vitro . Methods We used the model of primary neuron culture neonatal rats' cortex in vitro , measured the diameter of the cell and the length of the neurite,and observed the NSE and NGF expression in cultured neurons with immunohistochemical method.Results 1.The newborn rats' cerebral neurons could grow and develop in vitro . 2. Adding Se into the medium could increase the diameter of the cell and the length of the neurite of the cultured neurons in vitro . 3. Adding Se into the medium could promote NSE and NGF expression in comparison with control group.Conclusion The results indicate that selenium has an important biological significance in the neuronal growth, development and differentiation in early stage in vitro .
