党参皂甙对肾移植免疫排斥反应的作用

摘    要:目的研究党参皂甙对大鼠肾移植急性排斥反应的影响。方法采用原位大鼠肾移植模型,共设3个实验组:假手术组(SH)、生理盐水对照组(NS)、党参皂甙组(SA);根据观察时间点的不同又分别分3个亚组:3d组、5d组和8d组,于相应的时间点观察各组大鼠移植肾黏附分子ICAM-1的表达、受体脾脏IL-2的表达及移植肾排斥反应发生的程度。结果 IL-2和ICAM-1的表达,在术后5d时,对照组和皂甙组与假手术组相比,差异均有统计学意义(P<0.05),但对照组与皂甙组间差异无统计学意义(P>0.05);第8天时,各组间差异均有统计学意义(P<0.05),电泳条带亮度假手术组最弱,对照组较强,皂甙组最强。结论党参皂甙应用于肾移植可能加重移植肾的排斥反应,肾移植缺血-再灌注损伤期过后应慎用党参皂甙。

Abstract:Objective To study the role of codonopsis saponin in renal transplant immune rejection.Methods A model of renal transplantation in situ was used.All rats were divided into 3 groups: sham-operation group(SH),normal saline control group(NS) and codonopsis saponin intervention group(SA).Then according to the observation time points they were further divided into 3 different subgroups,respectively: 3 d group,5 d group and 8 d group.At the corresponding time points we observed the expression of each rat’s renal allograft adhesion molecules ICAM-1,the expression of receptor spleen IL-2 and the degree of renal allograft rejection of the renal allagraft.Results The expressions of IL-2 and ICAM-1 at day 5 after operation were significantly different in NS group and SA group from those in SH group(P<0.05),but NS group and SA group did not differ significantly(P>0.05).At day 8,significant differences were observed among all the groups(P<0.05);the brightness of electrophoresis strip was the weakest in SH group,stronger in NS group and the strongest in SA group.Conclusion Codonopsis saponin may increase renal allograft rejection;therefore,we had better stop using codonopsis saponin after renal transplant ischemia-reperfusion injury.
