低硒大骨节病病区粮对大鼠软骨Ⅱ型胶原合成代谢的影响

摘    要:为探讨病区粮及硒对软骨胶原代谢的影响,采用低硒大骨节病病区粮,病区粮加硒和非病区粮喂养大鼠66d,观察了低硒KBD病区粮和硒对软骨Ⅱ型胶原合成代谢的影响。结果表明:①病区组Ⅱ型胶原甘氨酸、丙氨酸等疏水氨基酸含量较非病区组增高,羟脯氨酸和羟赖氨酸含量降低。病区粮加硒后上述变化可得到部分纠正。②病区组软骨Ⅱ型胶原脯氨酸和赖氨酸羟化程度分别较非病区组降低6.4%和7.7%。加硒组较病区组脯氨酸羟化程度增高9.4%,但赖氨酸羟化程度未增高。③病区组Ⅱ型胶原电泳迁移率及对胰蛋白酶敏感性与其它组无显著差别。

Abstract:In the feeding trials rats were fed on the dietsof: (A) cereals from KBD endemic area (KBDgroup); (B) Se supplemented cereals from thearea as indicated above (Se-sup group); (C) cere-als from non-KBD endemic area (non-KBDgroup). The effects of cereals from KBD endemicarea and Se on cartilage collagen type Ⅱ synthesismetabolism were investigated. The resultsshowed: ①. In KBD group the amounts. of hy-drophobic amino acids such as glycine and alaninewere higher, while the amounts of hydroxyprolineand hydroxylysine were lower than those if non-KBD group. In Se-sup group the parameters men-tioned above rectified apparently. ②. The prolineand lysine hydroxylations of collagen type Ⅱ inKBD group decreased respectivly by 6. 4% and7. 7% when compared with those of non-KBDgroup. In Se-sup group the proline hydroxylationincreased by 9. 4%, but the lyisine hydroxylationwas unchanged compared with that of KBD group.③. the electrophoresis mobility and trypsine-sus-ceptibility at 37C of collagen type Ⅱ were not sig-nificantly different among three groups. These results indicated that cereals from KBDendemic area resulted in rats cartilage collagentype Ⅱ underhydroxylated, the amounts of hydrophobic amino acids increased, but there was noeffect on the stability of triple helices and molecu-lar weight of collagen type Ⅱ, and that Se-supple-mented cereals from KBD endemic area partialy im-proved the synthesis metabolism of cartilage colla-gen type Ⅱ.
