摘    要:目的 观察氟对牙胚细胞增殖及相关调控蛋白表达的影响。方法 采用人牙胚体外器官培养模型 ,组织学观察低浓度 (10mg·L- 1 )和高浓度 (2 5mg·L- 1 )氟对细胞增殖的影响 ;免疫组织化学及图像分析技术检测氟对细胞周期蛋白D1 (cyclinD1 )和细胞增殖核抗原 (PCNA)表达的影响。结果 氟处理组细胞增殖受抑 ,cyclinD1 和PCNA的表达显著降低 (P <0 .0 1)。结论 氟化物可能通过抑制cyclinD1 和PCNA的表达引起细胞增殖抑制 ,导致牙胚发育异常

Abstract:Objective To investigate the effect of fluoride on human tooth-germ cell proliferation and expression of related regulators. Methods By using an organ culture model of human tooth-germ in vitro, the effects of low (10?mg·L -1) and high (25?mg·L -1) concentrations of fluoride on cell proliferation were observed histologically and the expression of cyclinD 1 and proliferating cell nuclear antigen (PCNA) were detected by immunohistochemistry staining and image analysis technique. Results Fluoride inhibited cell proliferation and evidently decreased the expression of cyclinD 1 and PCNA. Conclusion Fluoride may inhibit tooth-germ cell proliferation by inhibiting the expression of cyclinD 1 and PCNA, which leads to the abnormal development of teeth.
