摘    要:目的 观察高脂饮食对正常大鼠胰岛素敏感性的影响。方法 用等热卡高脂饮食 (HF)饲养正常大鼠 ,并以胰岛素增敏剂曲格列酮进行干预 ,用葡萄糖 胰岛素耐量试验测定胰岛素敏感性 ,检测大鼠血压、血糖、胰岛素、内皮素 1、血脂、组织糖原及甘油三酯等指标。结果 等热卡HF对大鼠体重、血压、血糖无影响 ,胰岛素、甘油三酯、胆固醇及内皮素 1水平明显升高 ,高密度脂蛋白和组织糖原含量下降。HF 4周时已出现胰岛素抵抗 (IR) (K值 5 .6 3,与对照组K值7.77比较 ,P <0 .0 1)。HF 7周时 ,IR进一步加重。曲格列酮干预后 ,胰岛素敏感性明显改善。结论 HF能够诱发正常大鼠发生胰岛素抵抗 ,组织糖原合成下降与胰岛素抵抗有关

Abstract:Objective To observe the effects of high-fat diet on insulin sensitivity in rats. Methods Normal rats were fed with equicalorical high-fat diet and then treated with troglitazone. Glucose-insulin tolerance test was used to measure insulin sensitivity, blood pressure, blood glucose, insulin, endothelin, lipid, tissue glycogen and triglyceride levels. Results high-fat diet had no effect on body weight gain and blood pressure. Fasting and postprandial blood glucose levels were similar among the different groups. The levels of blood insulin, triglyceride, cholesterol and endothelin increased markedly, and the decrease in tissue glycogen level was also found. high-fat feeding caused insulin resistance after 4 weeks (K 5.63 vs. control 7.77, P<0.01). More evident insulin resistance could be seen after 7-week high-fat feeding (K 3.07 vs. control 7.71, P<0.01). Troglitazone could improve insulin sensitivity significantly. Conclusion High-fat diet can cause insulin resistance in normal rats, and the decreased tissue glycogen level might be related with insulin resistance.
