硒对胎鼠肝细胞核酸及蛋白质合成的影响

摘    要:本文报道了微量元素硒对培养纯种Wistar大白鼠胎肝细胞核酸及蛋白质合成的影响。观察了肝细胞培养基中不同硒水平对~3H-TdR(~3H-脱氧胸苷)、~(14)C-UR(~(14)C-尿苷)参入率的影响,同时还测定了培养基中白蛋白及甲胎蛋白(AFP)的含量。结果显示:细胞培养基中硒(Se)浓度在0.4~0.6μg/ml范围内使~3H-TdR、~(14)C-UR的参入率、白蛋白及甲胎蛋白含量均明显高于不补硒组。说明硒在这一浓度范围内能促进肝细胞DNA、RNA更新率及蛋白质合成。

Abstract:In order to determine the effect of trace elementselenium on dcoxyribonucleic acid (DNA), ribonucleicacid (RNA) and protein synthesis in culturing fetal rathepatocytes, the incroporation of ~3H-deoxythymidine(~3H-TdR) into DNA, ~(14)C-uridine (~(14)C--UR) intoRNA by fetal rat hepatocytes cultured inselenium--supplemented culture medium were investigated In addition the amounts of albumin and fetalprotein released from the same hepatocytes into the cul-ture medium were also determined. It was shown thatthe incorporation rate of ~3H--TdR into DNA and~(14)C--UR into RNA by fetal hepatocytes cultured in themedium supplemented with selenium as selenite at arang of Q 4 to Q 6 μg per ml levels were significantlyhigher than those cultured in medium withoutselenium--supplementation, concomitantly the amountsof albumin and--fetal protein released into the sameselenium--supplemented culture medium were alsogreater signicantly than those did in the mediumwithout selenium--supplementaion (as control). Theresults suggest that selenium supplementation in culturemedium at the levels of Q 4 to Q 6 μg selenium per mlcan promote the synthesis of DNA, RNA and proteinby fetal rat hepatocytes
