共刺激分子B_(7.1)拼接异构体的初步研究

摘    要:利用B1.7cDNA特异性引物、RT-PCR技术从新鲜淋巴瘤组织中扩增得到一与标准型B7.1片段长度不同的B7.1分子。进一步将该片段插入真核表达载体PSVL中,转染细胞得到了较高水平的表达。免疫组化研究结果证实抗B7.1单抗可与该片段编码的蛋白结合,提示该cDNA片段为B7.1抗原的拼接异构体。

Abstract:B7. 1 is the most important costimulatory molecule and has been studied extensively. We have got a B7.1 isoform molecule whose length was different from the normal B7.1 fragment by using RT-PCR technique in lymghoma biospies. This isoform was inserted a eukarvotic expressinq vector and trans fected into Hela cell line. The expressing situation was studied by immunohistochemistry technique using B7.1 monoclonal antibody. The results suggest that there is homogeneity, between the protein encoded by this isoform B7.1 molecule and prototype B7.1 protein.
