宫颈鳞癌中端粒酶活性与p53和C-myc的表达

摘    要:目的 探讨宫颈鳞癌中端粒酶活性、凋亡相关基因p5 3和C myc的表达水平及三者间关系。 方法 用PCRELISA法检测正常宫颈组织、宫颈上皮内瘤样变 (CIN)及浸润性宫颈鳞癌组织中端粒酶活性 ,用免疫组化ABC法检测 p5 3、C myc蛋白的表达。 结果 ①浸润性宫颈鳞癌中 ,端粒酶活性、p5 3、C myc蛋白表达水平明显增高 (P <0 .0 5 ) ;②端粒酶阳性率、p5 3和C myc蛋白表达水平与宫颈鳞癌分化程度有关 ,即肿瘤分化程度越差 ,端粒酶阳性率越高 (P <0 .0 5 ) ,而与宫颈鳞癌病理类型、临床分期无关 ;③宫颈鳞癌中端粒酶活性、p5 3表达、C myc表达三者之间有明显的内在联系 (P <0 .0 5 )。结论 ①端粒酶活性增高在宫颈鳞癌的发生及发展中起重要作用 ;②端粒酶活性、p5 3、C myc蛋白水平可作为宫颈鳞癌诊断和预后指标 ;③在宫颈鳞癌的形成和发展中 ,端粒酶与凋亡相关基因的表达可能具有协同作用

Abstract:Objective To investigate the relationship between telomerase and apoptosis associated oncoproteins?, and the role of them in the process of carcinogenesis and progression of cervical cancer. Methods PCR ELISA and immunohistochemical techniques were used to detect telomerase activity and the expression of p53, C myc oncoprotein in normal and abnormal cervical tissues, respectively. Results ① Telomerase activity, p53 and C myc protein levels were significantly increased in cervical cancer group ( P < 0.05 ); ② Lower level of differentiated degree in cervical cancer was related to the higher levels of telomerase activity, p53 and C myc protein expression; ③ The telomerase activity, the expression of p53 and C myc were correlated in invasive cervical cancer ( P < 0.05 ). Conclusion ① The telomerase activity, p53, C myc genes may play an important role in the process of carcinogenesis and progression of cervical cancer. ② The telomerase activity, p53, C myc can be used as useful markers of early diagnostic and prognostic factor in cervical cancer; ③ Telomerase activity is in coordination with apoptosis associated oncoproteins in the process of carcinogenesis and progression of cervical cancer.
