COX-2、VEGF的表达和胃癌血管生成的关系

摘    要:目的 探讨环氧化酶 2 (COX 2 )、血管内皮生长因子 (VEGF)与胃癌血管生成的关系。方法 用免疫组化SP法检测 4 5例胃癌、癌旁组织COX 2、VEGF和MVD的表达。结果 胃癌组织COX 2、VEGF和MVD的表达明显高于癌旁组织。COX 2 (+)或VEGF(+)组MVD(6 1.2 9± 14 .31;6 0 .0 7± 15 .86 )均显著高于COX 2 (- )或VEGF(- )组 (45 .38± 12 .4 2 ;4 3.0 6± 18.91,P <0 .0 5 ) ,COX 2、VEGF均为阳性组MVD(6 2 .4 6± 14 .34)明显高于两者均阴性组 (41.2 4± 13.6 5 ,P<0 .0 5 )或仅一项阳性组 (P <0 .0 5 )。COX 2、VEGF均与MVD显著正相关 (P <0 .0 1) ,COX 2与VEGF显著正相关 (P<0 .0 1)。结论 COX 2可能参与胃癌血管生成 ,VEGF可能是COX 2诱导肿瘤血管生成的重要介质

Abstract:Objective To investigate the expression of cyclooxygenase-2 (COX-2) and vascular endothelial cell growth factor (VEGF) in gastric carcinoma and its relation with the microvessel density (MVD). Methods The expressions of COX-2, VEGF and MVD of gastric carcinoma were studied by immunohistochemical method in 45 surgical resected specimens. Results The positive rates of COX-2,VEGF and MVD in gastric carcinoma were significantly higher than those in paracancerous tissues. The MVD in COX-2(+) (61.29±14.31)and VEGF(+) (60.07±15.86)tumors was significantly higher than that in COX-2(-) and VEGF(-) tumors (45.38±12.42; 43.06±18.91,respectively,P<0.05), and MVD (62.46±14.34,P<0.05)in both COX-2(+) and VEGF(+) tumors was significantly higher than that in the other subgroups. A close correlation was found between MVD and COX-2 (P<0.01), and VEGF expression was correlated with both COX-2 expression and MVD (P<0.01). Conclusion COX-2 expression might correlate with tumor angiogenesis in gastric carcinoma.It is likely that VEGF is one of the most important mediators of the COX-2 angiogenic pathway.
