SLE患者外周血单个核细胞分泌IL-4、IL-10、IL-12和IFN-γ水平及意义

目的探讨系统性红斑狼疮(SLE)患者外周血中Th1/Th2偏移现象。方法采用外周血单个核细胞(PBMC)培养和酶联免疫吸附试验(ELISA)检测37例SLE患者和17例健康人外周血单个核细胞(PBMC)培养上清液中IL-4I、L-10I、L-12和干扰素-γ(IFN-γ)水平。结果SLE患者的IL-10水平高于健康对照组(P<0.05);IL-12低于健康对照组(P<0.01);IL-4和IFN-γ与健康对照组相比无统计学意义(P>0.05)。结论SLE患者外周血Th1/Th2细胞平衡失调,IL-10升高,IL-12下降。

Level and significance of IL-4, IL-10, IL-12 and IFN- γ secreted by peripheral blood mononuclear cells in patients with systemic lupus erythematousus

Objective To investigate the imbalance phenomenon of Th1/Th2 cells of peripheral blood in patients with systemic lupus erythematousus(SLE).Methods The culture supernatant levels of IL-4,IL-10,IL-12,and interferon-γ(IFN-γ),that were produced by peripheral blood mononuclear(PBMC) were detected by Sandwich ELISA in 37 SLE and 17 healthy subjects.Results The level of IL-10 (230.94±309.14)ng/L] produced by PBMC from the patients of SLE was significantly higher than that of normal controls (85.58±91.87)ng/L(P<0.05)].The level of IL-12 (24.40±20.08)ng/L] was significantly lower than that of normal controls (43.53±25.14)ng/L];the level IL-4 (32.82±11.31)ng/L] and IFN-γ (263.80±255.18)ng/L] were not statistically different between SLE patients and controls IL-4 (27.95±7.55)ng/L,INF-γ(380.61±299.13)ng/L,P>0.05].Conclusion There is imbalance of Th1/Th2 cells in patients with SLE: level of IL-10 was higher while level of IL-12 was lower than that in normal controls.
