摘    要:目的 探索雪腐镰刀菌烯醇 (NIV)对培养软骨细胞DNA损伤作用及硒的保护作用。方法 应用软骨细胞单层培养、碱性单细胞微量凝胶电泳、琼脂糖电泳和生化方法检测软骨细胞DNA量及损伤情况。结果 在实验设定的NIV浓度范围内 ,随NIV浓度增加DNA含量减少、DNA受损细胞增多和DNA损伤加重 ,同浓度NIV加硒组损伤情况好于未加硒组。结论 NIV对培养软骨细胞有明显损伤作用 ,加硒有保护作用。

The effect of nivalenol and the protection of selenium on DNA of cultured chondrocytes
Abstract:Objective To investigate the effect of nivalenol(NIV) and the protection of selenium(Se) on DNA of the cultured chondrocytes. Methods The DNA contents in the single layer cultured chondrocytes were assessed by the biochemical methods,and the DNA damages were observed by the single cell microgel electrophoresis assay and the agarose gel electrophoresis assay.Results In the media containing different concentrations of NIV (0.0005~0.0200?mg/L),the DNA contents were decreased and the DNA damages were worsen with the concentration of NIV in the given does.When Se was added into the media,the DNA damages by NIV were decreased,but not completely.Conclusion It caused evident damages to the cultured chondrocytes when NIV concentration is the concentration of this experiment. Se can prevent the DNA damage.
