丙型肝炎病毒核心蛋白基因序列变异分析

摘    要:为了研究西安地区丙型肝炎病毒(HCV)基因组中C区基因的变异性,采用单链构象多态性(Single-StrandConformationPolrmorphism,SSCP)分析万法,对36份HCVRNA阳性血清的PCR产物进行分析,结果有31份样品的SSCP图谱相同,对其中二份样品进行测序,证明序列是相同的;另外5价样品SSCP图谱不相同,其序列与前者也有差异。说明本地区HCV基因组中C区基因存在一个主要的基因类型。该实验为本地区HCV诊断试剂和疫苗的研究确定了研究材料。

Abstract:in order to research the variability ofHCV core protein gene (HCV C gene) in Xi'an, weanalyzed 36 pieces of serum's PCR products of HCVgene by SSCP. The results showed that 31 sampleshad the same SSCP graph. We sequenced 2 PCRproducts of the 31 and found out that their se-quences were the same. The other 5 samples' SSCPgraphs were different from the 31 samples. We sequenced one PCR product of the 5 and the resultwas different from the 2 samples' we had sequenced. It proves that there is a main type of HCVC gene in Xi'an. The experiment points out a way toresearch the diagnosis kit and vaccine of HCV inXi'an.
