奥曲肽对人肝癌细胞株MHCC97-H的影响

目的探讨奥曲肽对人高转移潜能肝癌细胞株MHCC97-H的影响及其机制。方法奥曲肽体外作用于生长对数期人高转移潜能肝癌细胞株MHCC97-H,观察生长曲线的变化;应用MTT比色分析法分析细胞的生长抑制作用;透射电镜下观察细胞形态变化。结果奥曲肽在0.2 mg/L质量浓度下体外干预MHCC97-H细胞使生长受抑;透射电镜下观察呈现凋亡形态改变;在0.05-0.8 mg/L质量浓度范围内呈剂量依赖关系抑制人高转移潜能肝癌细胞株MHCC97-H增殖(P<0.01),并出现饱和现象。结论奥曲肽对人高转移潜能肝癌细胞株MHCC97-H增殖具有抑制作用,在一定范围内呈剂量依赖关系,其机制与诱导肿瘤细胞凋亡有关。

Effect of Octreotide on human hepatocellular cancer cell line MHCC97-H in vitro

Objective To study the effect of Octreotide on human hepatocellular cancer cell line MHCC97-H and its mechanism. Methods Human hepatocellular cancer cell line MHCC97-H was treated with Octreotide in vitro,growth curve was observed,and cell proliferation was detected by MTT assay.Apoptosis was confirmed by transmission electron microscope.Results Human hepatocellular cancer cell line MHCC97-H treated with Octreotide at 0.2mg/L level was inhibited and apoptosis cells were found under transmission electron microscope.The proliferation of MHCC97-H was inhibited significantly by Octreotide in a dose-dependant manner(ranging from 0.05 to 0.8 mg/L,P<0.01).Conclusion Octreotide can inhibit the proliferation of human hepatocellular cancer cell line MHCC97-H and the effect appears to be dose-dependant;the mechanism is related to inducing apoptosis.
