大鼠背海马CCK样神经元的衰老变化——ABC法和图像定量分析研究

摘    要:本文采用免疫组化ABC法和图像定量分析研究,观察了不同年龄组大鼠背海马内胆囊收缩素(CCK-8)样神经元的衰老变化:CCK样神经元见于背海马的所有区和层,主要分布于海马本部的锥体层,放射层,腔隙分子层和齿状回的多形层。除齿状回CCK样神经元数从幼年到老年一直持续缓慢增加外,其余各区表现为中年组CCK样神经元数显著性增加,老年时神经元数又减少;CCK样神经元胞体截面积呈增龄性持续增大,且各区,层的增大程度不均衡;背海马CCK样神经元灰度值呈增龄性升高,但老年时升高的速度减慢;CCK样神经元形态在老年表现为胞体边缘僵硬,胞浆染色不匀,突起变粗伸长,分枝增多。

Abstract:The aging changes of CCK--8--like neurons inthe dorsal hippocampus of the rats (2, 11, 24months)were studied with the immunohistochemistry(ABC)method and image quantitative analytical study.CCK--like neurons were located at all regions and lay-ers in the dorsal hippocampus, but it was relativelymore abundant in stratum pyramidale, stratumradiatum, stratum lacunosum--moleculare and thepolymorphous layer of dentate gvrus. Except thatCCK--like neuron number in dentate gyrus increasedslowly from the young to the old, CCK--like neuronsof other regions inercased significantiy in the adult, to-ward the old there was a decrease of the ncurons. Theaverage square of CCK--like ncurons increased withaging increasig extent of regions and layers varied eachother. The grey value of CCK--like neuronsascended continously with aging but ascendant ratc inthe old declined From the young to the old themorphological changes of the neurons appeared thestiffness of perikaryon outlines, and the unevenness ofcytoplasma staining. The processes became larger andlonger. Meanwhilc, the number of the branch increased.
