摘    要:目的 研究全反式维甲酸 (ATRA)和干扰素 γ(IFN γ)联合应用对胶质瘤细胞增殖、凋亡及侵袭力的协同影响。方法 ATRA和IFN γ共同处理人胶质瘤细胞系SHG 4 4 ,MTT比色法检测细胞增殖能力 ,流式细胞仪检测瘤细胞周期分布和凋亡率 ;根据瘤细胞穿过Matrigel胶的能力 ,检测瘤细胞体外侵袭力 ;以ATRA、IFN γ单独处理的细胞作为对照。结果 胶质瘤细胞经ATRA和IFN γ联合处理后增殖能力下降 ,凋亡率增加 ,细胞阻滞于G1期 ,其侵袭性降低 ,与单独处理组相比较差异有显著性 (P <0 .0 5 )。结论 ATRA和IFN γ联合应用具有协同抑制人胶质瘤细胞系SHG 4 4恶性表型和诱导凋亡的作用

Abstract:Objective To investigate the synergistic effect of all-trans-retinoic acid (ATRT) and interferon-γ(IFN-γ) on the proliferation, apoptosis and invasion of human glioma cell line SHG-44. Methods The glioma cellular proliferation capacity was observed with MTT assay, the cell cycle distribution and the apoptotic rate were measured with flow cytometry, and the invasion capacity was assayed in a Transwell cell culture chamber. Results The proliferation capacity and invasion capacity decreased, and the apoptotic rate and percentage of cells in G 1 phase increased in the glioma cells treated by combination of RATA and IFN-γ. Conclusion RATA and IFN-γ combined have synergistic effects in inhibiting glioma cells and inducing the apoptosis.
