摘    要:宫颈癌的流行病学特点提示癌的发生与感染因素有关。主要涉及两组病毒:单纯疱疹病毒(HSV)和人乳头瘤病毒(HPV)。为了探讨HSV、HPV 与宫颈癌的关系,我们以~(32)P 标记HSV—2DNA 和HPV DNA,分别用Southern 转印和打点杂交技术,同时检测了73例宫颈癌和14例正常宫颈活检标本。在严格状态下杂交,65例宫颈癌中未发现HSV—2DNA,其中19例癌组织RNA 与HSV—2DNA 杂交阳性。研究发现与宫颈癌组织RNA 杂交阳性的HSV—2DNA 位于0.2~0.33,0.58~0.625图谱单位。此外,在25例宫颈癌中检出了HPV_(16) DNA,2例标本检出了HPV_(18) DNA。在非严格状态下杂交,8例经PstⅠ酶切的宫颈癌组织DNA,Southern 转印后分别与HPV_(16) DNA 和HSV—2BglⅡ克隆片段杂交。结果,6例检出了HPV_(16)DNA,1例既检出了HPV_(16)DNA,又检出了HSV—2DNA。初步结果说明HPV、HSV—2与宫颈癌的发生可能有关。

Abstract:Epidemiological studies suggsted thatthe etiology of cervical cancer was closelyrelated to some infectious agents,mainlyinvolving two groups of virus:Herpes Si-mplex Virus(HSV)and Human Papillomavirus(HPV).In order to study the relati-onship between HSV,HPV and cervicalcancer,we surveyed 73 cervical cancer bi-opsies and 14 normal cervix biopsies byusing ~(32)P—labeled HSV-2DNA and ~(32)P—labeled HPVs DNA as probes and part byusing Southern transfer and spotblot hybr-idization.Under stringent condition,none of 65cervical cancer samples were found to con-tain HSV-2 sequences,but 19 extractedRNA gave positive results when hybridza-tion with HSV-2 DNA.After the researchwe detected that HSV-2 DNA hybridizedwith cellular RNA located at 0.2 to 0.33map unit and 0.58 to 0.625 map unit.HPV 16 DNA were detected in 25 cases ofcervical cancer and HPV 18 DNA weredetected in 2 cases.Under non-stringentcondition,8 cellular DNA extracted fromcervical cancer were analysed by Southerntransfer hybridization after cleavage withrestriction endonuclease Pst Ⅰ,using~(32)P-labeled HSV-2 DNA and ~(32)P-labeled HPV16 DNA as probes.6 samples were foundto contain HPV16 sequences and I caseto contain both HPV and HSV-2 sequences.It suggested that HPV and HSV-2 mayplay important roles in cervical cancer.
