镓盐和雌二醇对维甲酸致骨质疏松大鼠骨代谢的影响

摘    要:
目的 对比研究镓盐及雌二醇对维甲酸致大鼠骨质疏松模型骨代谢的影响。方法  3月龄SD雌性大鼠 45只 ,随机分为两组 :正常组 ( 1 3只 ) ,骨质疏松组 ( 32只 ) ,骨质疏松组按 85mg/(kg·d)维甲酸灌胃 1 5d。然后进入治疗阶段 ,实验分为四组 :正常组 ( 8只 ) ,骨质疏松组 ( 9只 ) ,正常喂养 ;氯化镓治疗组 ( 1 0只 ) ,2 5mg/(kg·d)氯化镓灌胃 ;雌激素治疗组( 8只 ) ,雌激素 0 .2 μg/kg ,3次 /周 ,腹腔注射 ,治疗两个月。 结果 模型期骨质疏松组血清AKP和TRAP活性显著增高 ,骨质疏松组平均骨小梁宽度变窄 (P <0 .0 0 1 ) ,平均骨小梁间距变大(P <0 .0 0 1 ) ,平均骨皮质厚度变薄 (P <0 .0 0 1 ) ,骨小梁百分比降低 (P <0 .0 0 1 ) ,镓盐及雌激素治疗后 ,可使显著增高的AKP和TRAP活性降低到接近正常组水平 ,平均骨皮质厚度增加 ,骨小梁百分比增大。结论 雌激素抑制骨吸收 ,促进骨形成 ,维持骨转换平衡。提示镓盐通过抑制破骨细胞的破骨能力 ,降低了骨转换率 ,抑制骨溶解 ,增加了骨量及骨的硬度。

Experimental study on effect of gallium salt and estradiol on bone metabolism in tretinoin-induced osteoporosis of rats
Abstract:
Objective To investigate the influence of gallium salt and estradiol on bone metabolism in tretinoin induced osteoporosis of rats. Methods Forty five Sprague Dawley (SD) female rats,three months of age,were randomly divided into control and osteoporosis group with 85?mg/(kg·d) for 15?d.Then we divided them into control group,osteoporosis group,gallium chloride treated group (25?mg·kg -1 ·d -1 ),estrogen treated group (in dosage of 0.2?μg/kg and 3 times per week, treated for 2 months).Results After duplicating osteoporosis model,rat's activities of AKP and TRTP increased significantly,the trabecular bone volume,trabecular thickness and the thickness of bone cortex reduced significantly at the same time,the space between trabecular bone largened significantly. Two months after treatment with gallium salt and estrogen,AKP and TRAP decreased to the level of to control group closely.Trabecular thickness and the thickness of bone cortex increased significantly.Conclusion Estrogen restrain bone resorption,advance the formation of bone,keep the balance bone tumover.Gallium salt restrain the function of osteroclast,decrease the rates of bone tumover,inhibit bone dissolution,increase the quantity and rigidity of bone.
