大鼠钴中毒心肌病和过氧化反应关系的研究

摘    要:本文对腹腔注射氯化钴(每日5mg/kg)大鼠心肌病和过氧化反应的关系进行了研究。在注射钴次3和5次后血浆磷酸肌酸激酶和谷草转氨酶活性增高。注射钴7次后,心肌脂质染色增强,部分动物心脏显示形态学异常和琥珀酸脱氢酶反应减弱;心肌游离脂肪酸和血清维生素 E 含量增加.心肌硒含量和超氧物歧化酶活性均无改变。心肌谷胱甘肽过氧化物酶活性在注射钴3次和5次后也无变化;尽管在注射钴7次后其活性下降,但心肌脂质过氧化物水平并不升高。结果表明钴对心肌的损害作用与脂质过氧化物的形成无明显关系.

Abstract:The present studies were designed to investigate the cardiomyopathy produced in rats by the administration of cobalt in terms of its relation to lipidperoxidation and protective agents involved in peroxidative pathways.48 Wistar rats weighing 100 gwere divided into cobalt(Co)and control groups of 24rats each,and supplied with a stock diet adlibitum.The Co group received ip injections of 5 mg/kgbody wt daily for 3 days,and then every two days 4times.The control was given the same volume of sa-line injection.Each group was subdivided into 3subgroups of 8 rats each,and rats were killed post3,5and 7 injections of Co,respectively.The Co-treated rats,as compared to the control,showed significant increases in plasma CPK and GOTafter 3 and 5 injections of Co.Under light microscope,the heart in part of the rats after 7 doses had remarka-ble cardiac lesion.Histochemically,thesuccino-dehydrogenase was less intense and lipidstaining was increased (?) intensity.Marked increases incardiac free fatty acid and seru itamin E contents were seen.No significant changes for cardiac selenium content and superoxide dismutase activity wereobserved.Cardiac glutathione peroxidase activities wereunchanged after 3 and 5 doses.Although the activity de-creased after 7 injections,there was no alteration incardiac lipid peroxide level.The results demonstratethat the Co-induced cartdiomyopathy is not related toa formation of lipid peroxide.
