硒、蛋氨酸对饲克山病病区粮大鼠心、肝组织溶酶体膜的影响

摘    要:利用差迷离心法分离提取大鼠心、肝组织富含溶酶体部分,并以溶酶体之标志酶酸性磷酸酶(ACP)及组织蛋白酶D(CD)的游离酶和总酶活性比值为观察溶酶体膜稳定性变化的指标。分别观察了低硒的克山病病区粮及病区粮加亚硒酸钠或蛋氨酸喂养的大鼠之心、肝溶酶体酶(ACD和CD)的活性变化。发现用低的病区粮喂养的大鼠心肝组织溶酶体膜的稳定性下降,而加硒或蛋氨酸后,其稳定性增加。考虑这种变化与大鼠心肝组织中的GSH-P_x活性变化有关。

Abstract:Free enzyme(FE) and total enzyme(TE)activities of acid phosphatase(ACP) and cathepsin D(CD)of the heart and liver extracts were determined by feeding 4 groups of rats. The Se content in feed was 0.01 ppm in the gronp of the Keshan disease endemic area(Group Ⅰ), 0.14 ppm in the seleninm-supplemented group(Group Ⅱ), 0.01 ppm in the methionine-supplemented group(Group Ⅲ), and 0.20 ppm in the Xi'an group(Group Ⅳ), respectively. In addition, the GSE-Px activities of the heart and liver and those of malonaldnyde(TBA) in the heart were determined. The results show: 1 ) The FE and TE activities of ACP and CD in the heart and liver and the ratio of FE to TE were significantly higher in Group Ⅰ than in Group Ⅳ. TBA in the heart was increased in Group Ⅰ, but the values of GSE-Px in the heart and liver were lower in Group Ⅰ than in Group Ⅱ. 2) The FE and TE activities and the ratio of FE to TE in Groups Ⅱ and Ⅲ were nearly as normal as in the contral group and so were the GSH-Px and TBA values. It is suggested that Se difficiency may affect the lysosomal stability and the peroxidative damage to the lysosomal monbrane in the rat and that the feed supplemented with selenium or methionine has protective actions on the lysosonal menbrane, resulting in the increased its stability against the peroxidative damage.
