鼻咽癌组织中BCRF-1基因表达及其与免疫功能的相关性分析

摘    要:目的研究鼻咽癌组织中BCRF-1mRNA及病毒白介素-10(VIL-10)蛋白质的表达,明确VIL-10对鼻咽癌患者免疫细胞的影响。方法在鼻咽癌组织和鼻咽炎症组织中,分别应用RT-PCR及Western Blot检测BCRF-1mRNA和蛋白产物VIL-10的表达;流式细胞术检测两组患者外周血CD4+T、CD8+T细胞的比例。结果①BCRF-1mR-NA在34/40例(85%)鼻咽癌组织和16/20例(80%)鼻咽炎症组织中表达阳性,两者差异无统计学意义(P>0.05)。②30/40例(75%)鼻咽癌组织VIL-10呈阳性,10/20例(50%)鼻咽炎症组织VIL-10呈阳性,两者差异具有统计学意义(P<0.05)。③40例鼻咽癌患者分为VIL-10阳性组和VIL-10阴性组,VIL-10阳性鼻咽癌患者组CD4+T细胞及CD4+T/CD8+T比值明显低于VIL-10阴性鼻咽癌患者组(24.99%±3.88%vs.35.92%±6.31%,0.86 vs.1.99,均P<0.05),而VIL-10阳性鼻咽癌患者组CD8+T细胞明显高于VIL-10阴性鼻咽癌患者组(30.61%±3.34%vs.20.10%±7.68%,P<0.05)。结论鼻咽癌组织中高表达BCRF-1基因及其蛋白产物VIL-10;VIL-10使鼻咽癌患者的免疫细胞发生偏移,发挥免疫抑制作用。

Abstract:Objective To investigate the expressions of BCRF-1mRNA and VIL-10 protein so as to clarify the effect of VIL-10 on immune cells of nasopharyngeal carcinoma(NPC).Methods The mRNA of BCRF-1 in tissues of NPC was detected by RT-PCR and VIL-10 protein was detected by Western blot.Flow cytometry was used to calculate the ratio of CD4+ T cells to CD8+ T cells.Results ①The mRNA of BCRF-1 was positive in 34 cases of NPC(34/40,85%) and 16 cases of nasopharyngeal inflammation(16/20,80%),without any significant difference between the two kinds of tissues(P>0.05).② VIL-10 protein was positive in 30 cases of NPC(30/40,75%) and 10 cases of nasopharyngeal inflammation(10/20,50%),with significant differences between the two kinds of tissues(P<0.05).③ Forty NPC patients were divided into VIL-10 positive and negative groups.The number of CD4+T cells and ratio of CD4+T/CD8+ were lower in VIL-10 positive group than in VIL-10 negative group(24.99±3.88% vs.35.92±6.31%;0.86 vs.1.99,P<0.05).The number of CD8+ T cells was greater in VIL-10 positive group than in VIL-10 negative group(30.61±3.34% vs.20.10±7.68%,P<0.05). Conclusion BCRF-1 gene and its protein product of VIL-10 have high expressions in tissues of NPC.VIL-10 can reduce the number of immune cells in NPC,which will cause immunosuppression.
