丹红注射液对U937细胞SR-AⅠ及ABCA1 mRNA表达的影响

摘    要:目的 以人单核细胞株U937细胞为研究对象,观察不同浓度丹红注射液对U937细胞清道夫受体AⅠ(SR-AⅠ)和ATP结合盒转运体A1( ABCA1)mRNA表达的影响,探讨其抗动脉粥样硬化的机制.方法 以100 nmol/L佛波酯诱导U937细胞48 h使其分化为巨噬细胞,再以50 mg/Lox-L DL(氧化低密度脂蛋白)孵育细胞的同时加入不同浓度丹红注射液或其他干预因素处理24 h,用实时荧光定量PCR方法检测其SR-A Ⅰ和ABCA1 mRNA的表达量.结果 与50 mg/L ox-LDL组比较,LXR-a(肝X受体激动剂)(T-090137)干预组U937细胞SR-A ⅠmRNA表达下降、ABCAl mRNA表达升高,但SR-A Ⅰ mRNA的表达无统计学意义(P>0.05),而ABCAl mRNA的表达有统计学意义(P<0.01);3.0 mL/L丹红注射液干预组U937细胞SR-A Ⅰ mRNA的表达升高,10.0、30.0、60.0 mL/L丹红注射液于预组U937细胞SR-A Ⅰ mRNA的表达逐渐下降,但各浓度组均无统计学意义(P>0.05);丹红注射液干预组ABCAl mRNA的表达则呈浓度依赖性升高,但于60.0mL/L浓度时出现表达下降,3.0、10.0、30.0 mL/L干预组有统计学意义(P<0.05).与LXR-a(肝X受体激动剂)(T-090137)干预组比较,丹红注射液各浓度干预组ABCAl mRNA表达降低,10.0、30.0 mL/L丹红注射液干预组ABCA1 mRNA的表达无统计学意义(P>0.05).结论 丹红注射液各浓度组对50 mg/Lox-LDL干预的U937细胞SR-A Ⅰ mRNA的表达无影响,而明显增加U937细胞ABCA1 mRNA表达,尤其在浓度10.0、30.0 mL/L时增加ABCA1 mRNA表达明显.增加U937细胞ABCA1 mRNA表达可能是其抗动脉粥样硬化的重要机制.

Abstract:Objective To study the effects of Danhong injection on expressions of SR-A I and ABCA1 mRNA in U937 cells and its antiatherosclerotic mechanism.Methods Human U937 cells were induced by phorbol 12-myristate 13-acetate for 48 hours,and then incubated with 50 mg/L ox-LDL for 24 hours with different concentrations of Danhong injection or other treatments.SR-AⅠ and ABCA1 mRNA expressions were detected by fluorescence quantitative PCR.Results Compared with 50 mg/L ox-LDL group,SR-AⅠ mRNA expressions of U937 cells treated by LXR agonist decreased and that of ABCA increased.However,SR-AⅠ mRNA expression had no significance(P>0.05),but ABCA1 mRNA expression was remarkable(P<0.01).The mRNA expression of SR-AⅠ treated by 3.0 mL/L Danhong injection increased,and then decreased gradually with the increase of Danhong injection concentration,but without significant differences among different concentration groups(P>0.05).ABCA1 mRNA expression of U937 cells treated by Danhong injection was concentration-dependently increased,but decreased in concentration of 60.0 mL/L;it had significance in different concentration groups except 60.0mL/L group(P<0.05).Compared with that in LXR agonist group,the mRNA expression of ABCA1 decreased in groups of different Danhong injection concentrations,no significant difference in 10.0mL/L or 30.0mL/L concentration groups(P>0.05). Conclusion Danhong injection of different concentrations does not affect SR-AⅠ mRNA expression in U937 cells treated with 50mg/L ox-LDL.It significantly increases the expression of ABCA1 mRNA in concentrations of 10.0mL/L and 30.0mL/L.The important antiatherosclerotic mechanism of Danhong injection may be related to the increased expression of ABCA1 mRNA.
