绞股蓝总皂甙对P388细胞的作用及机制探讨

摘    要:用单层平皿软琼脂克隆法证明绞股蓝总皂甙对体外培养的P388细胞克隆生长有明显的抑制作用,其IC50为(22.5±1.0)mg·L-1。为进一步探讨其抑制P388细胞生长机理,用放射性同位素掺入法观察了绞股蓝总皂成对P388细胞DNA、RNA及蛋白质合成的影响。结果表明,在绞股蓝总皂甙作用24h内,其主要抑制P88细胞DNA合成,轻度抑制RNA合成,而对蛋白质合成几乎无影响。

Abstract:Gypenosides (GP) inhibited cell growth of leukemia P388 in vitro by using cellular colony-forming assay method. The 50% inhibitory concentration of cell growth was (22. 5±1. 0)mg'L-1 respectively. In order to studv its inhibitory mechanism, radioactive isotope was incorporated in GP-supplemented 24 hours P388 cells. The results showed that GP mainly decreased DNA synthesis of P388 cells, slightly inhibited RNA synthesis and al-most had no effect on protein synthesis.
