诊断脑囊虫病快速ELISA试验方法建立与常规ELISA比较

通过提高抗原抗体结合反应的孵育温度(41℃),以缩短孵育时间,建立起检测脑囊虫病血清抗体的快速 ELISA 试验方法。在此温度下,血清抗体的几何平均滴度为812.58,较之在39℃、43℃和45℃条件下明显增高,同时阳性和阴性血清的免疫比值也最高,表明41℃是囊虫抗原抗体结合反应的适合温度。在此温度下,抗原抗体结合反应的速度明显增快,经过15分钟即可达到充分结合。用快速 ELISA与常规 ELISA 两种方法对30份脑囊虫病人血清同步测试结果表明,快速 ELISA 的光吸收值普遍略高于常规 ELISA。这就表明,在脑囊虫病的血清抗体检测中,快速 ELISA 完全可取代常规 ELISA,它有着简便、快速、敏感性高等优点。

THE ESTABLISHMENT OF FAST ELISA FOR DIAGNOSIS OF CEREBRAL CYSTICERCOSIS AND THE COMPARISON WITH ROUTINE ELISA

An experimental method of fast ELISA for determing serum antibody of cerebral cysticercosis was established by raising the incubating temperature (41℃)of antigen- antibody combining reaction so as to shor- ten the iecubating time (15 minutes).At the temperature of 41℃ the geometric av- erage titre of the serum antibody was 812.58,which was distinctly higher than at 39,43,and 45℃ temperature conditions. The immune ratio of positive with negative serum was the highest at this condition. It was indicated that the most appropriate temperture of cyst icercus,antigen-antibody reaction was 41℃.At this temperature, the velocity of the antigen-antibody combi- nation increases obviously.The combining reaction can reach a peak in 15 minutes. 32 patients′ serum of cerebral cysticercosis was determined by means of fast ELISA and the routine ELISA simultaneously. The results show that the optic absorbent value of fast ELISA was higher slightly titan that of routine ELISA,indicating that fast ELISA could completely replace routine ELISA in determing the serum an- tibody of cerebral cysticcrcosis,This me- thod has a lot of advantages,such as fast,high sensitivity etc.
