巨细胞病毒感染诱导移植排斥反应的机制研究

目的以人脐静脉内皮细胞(HUVECs)ECV304株作为研究对象,探讨巨细胞病毒(CMV)感染诱发排斥反应的机制。方法体外培养ECV304,施加不同干预因素,分为正常对照组、灭活病毒作用组、病毒作用组、病毒上清液作用组及病毒与更昔洛韦组。运用免疫组化技术检测不同组别内皮细胞表面细胞间粘附分子-1(ICAM-1)的表达。结果正常对照组内皮细胞ICAM-1的表达呈弱阳性,加入灭活的病毒及病毒上清液后,ICAM-1的表达仍为弱阳性(P>0.05);加入有活力的病毒后,内皮细胞ICAM-1的表达呈强阳性,积分指数显著高于上述3组(P<0.01);在CMV感染内皮细胞的同时加入更昔洛韦,细胞表面ICAM-1表达强度较病毒组并无减低(P>0.05)。结论CMV感染的内皮细胞表面ICAM-1的表达明显增加。通过诱导内皮细胞表面ICAM-1表达上调可能是CMV诱发排斥反应的机制之一。

Role of cytomegalovirus infection in renal allograft rejection: study of a possible mechanism

Objective To explore the mechanism of cytomegalovirus(CMV) induced the rejection reaction by observing the expression of the adhesion molecules on the surface of endothelial cell infected by CMV. Methods Endothelial cell strains ECV304 were cultured in vitro and influenced by various factors.The strains were divided into 5 groups,including normal control group,inactivated CMV infected group,CMV infected group,supernatant of CMV infected group and CMV combined ganciclovir group.The immunohistochemistry technique was used to detect the expression of ICAM-1 on the surface of endothelial cells.Results The expression of ICAM-1 on the surface of endothelial cells was weakly positive.After adding inactivated virus and supernatant of virus,it was still weakly positive.There was no significant difference(P>0.05);but adding active virus,it was strongly positive;the results were obviously higher than those of the other 3 groups,respectively(P<0.01).It indicated that CMV could increase the expression of ICAM-1 on the level of protein.Adding ganciclovir when the CMV infected endothelial cells,the results showed that the degree of intensity didn't decrease(P>0.05).Conclusion When infected with CMV,the expression of ICAM1 increased obviously.CMV caused rejection reaction mainly by inducing the increase of the expression of ICAM-1 in endothelial cells.
