五氯硫酚的毒性研究

为安全性评价提供毒理学资料对五氯硫酚用不同种属动物进行了多项毒性试验。结果表明，其经口ＬＤ５０雄性大鼠为９０９０ｍｇ／ｋｇ，而对小鼠大于１０ｇ／ｋｇ，属低毒物质。其蓄积系数大于５，故对小鼠无明显蓄积毒性。对家兔皮肤粘膜刺激作用轻微，但对豚鼠具轻度皮肤致敏作用。Ａｍｅｓ试验及微核试验中未显示诱变性。９０ｄ喂养试验中，中、低剂量组在血象、尿检、生化、病理及生长发育各观察指标未见异常改变，而高剂量组雄性大鼠的总进食量与体重增长明显低于对照，提示亚慢性毒作用阈剂量为９０９ｍｇ／ｋｇ。本研究未证实五氯硫酚具有卟啉原性作用。

STUDIES ON TOXICITY OF PENTACHLOROTHIOPHENOL

A series of toxicity tests were conducted on pentachlorothiophenol(PCTP)in labotatory animals of different species to provide toxicologic profiles for its safety assessment. The results of acute toxicity tests indicated that its oral LD50 values were 9090 mg/kg in male rats and more than 10 g/kg in mice, so it was categorized as a low toxic substance. The accumulative coefficient was more than 5 which means that the compound actually has no accumulating toxicity to mice. And also there were no obvious irritative effects of PCTP on either the skin or the mucousa sbown in rabbits,but thecompound was found to be a weak sensitizer in the guinea pig according to the results of GPMT of dermal sensitization. With both salmonella/microsome assay and micronucleus test in mouse bone marrow cells,PCTP gave negative results under the condition of the study.In the 90-day subchronic oral toxicity study,as compared with those in the control group, the total food intake and the weight gain of the male rats in the high dose group(909 mg/kg )were significantly reduced at the end of the dosing period, and no unusual changes were observed in bloOd picture, uninalysis, biochemical and pathologic examination, and growth and development for the medium and the low dose groups,Accordingly,the subchronic threshold dose of PCTP was estimate as 909 mg/kg.Moreover,no evidence of porphyrinogenic effect of PCTP was obtained in this study.
