人工冬虫夏草菌粉提高大鼠肝脏保存质量的实验研究

摘    要:目的 探讨人工冬虫夏草菌粉提取液对大鼠肝脏低温保存损伤的保护作用。方法 将一定剂量的虫草提取液加入乳酸林格液内,分别低温保存大鼠肝脏6、12 h后,再灌注30 min。通过高压液相色谱法(HPLC)检测肝脏组织内的ATP、ADP、AMP含量,测定再灌注流出液内的AST、ALT、MDA及ET含量。并切取肝脏标本,光镜及电镜观察肝组织损伤情况。结果 于保存6 h后,实验组AST、ALT、MDA和ET均低于对照组,但无统计学显著差异(P>0.05)。光镜下除轻度细胞水肿外,无其他明显病理改变;保存12 h后,实验组流出液ALT、MDA及ET含量明显低于对照组(P<0.05),肝脏组织内ATP、TAN及AEC显著高于对照组(P<0.01)。实验组细胞凋亡指数显著低于对照组(P<0.01)。实验组和对照组在光镜及电镜下的超微结构改变亦有明显差异。结论 人工冬虫夏草菌粉提取液可以显著提高保存肝脏的质量。

Abstract:Objective To investigate the protective effect of the extractions of synthetic Cordyceps Sinensia Sacc powder on cold preserved injured tissues of rat livers. Methods The extractions of synthetic Cordyceps Sinensia Sacc powder were added into LR at certain proportion. Having been preserved for 6 or 12 hours, the liver grafts of rats were reperfused for 30 minutes. And then, ATP, ADP, AMP in livers were measured and added up to total adenine nucleotide (TAN). Besides, AST, ALT, MDA, and ET in the effluent of the reperfusion were assayed. Pathological sections were studied and apoptosis index was detected by TUNEL method. Results The values of ATP, TAN in the experimental group preserved for 12 hours were significantly higher than the control group (P<0.01), while the changes of the values in the effluent of ALT, MDA, and ET were just opposite (P< 0.05 ). As for the experimental group and the control group preserved for 6 hours, the values of AST, ALT, MDA, and ET were of no statistical difference (P>0.05), but the mean of the former was higher than the latter. There was obvious increase of apoptosis index (P<0.01) between the experimental group and the control group preserved for 12 hours. Remarkable pathological difference between them was observed through light microscopy and electron microscopy. Conclusion Manual Cordyceps Sinensia Sacc powder can notably improve the quality of cold preserved rat livers.
