乳腺癌中COX-2、HER-2的表达及其相关性

摘    要:目的 研究环氧化酶 2 (COX 2 )和表皮生长因子 2 (HER 2 )在乳腺癌中的表达及其相关性。方法 应用免疫组织化学方法检测了 80例乳腺癌、2 7例乳腺不典型增生以及 2 0例正常乳腺组织中COX 2和HER 2表达 ,分析其与临床病理的相关性。结果 乳腺癌组织中COX 2阳性表达率为 5 7.5 % (4 6 / 80 ) ,COX 2表达与淋巴结转移、临床分期、组织学分级、雌激素受体相关 (P <0 .0 5 )。 37.5 % (30 / 80 )乳腺癌中HER 2阳性表达 ,HER 2表达与临床分期、淋巴结转移、组织学分级、雌激素受体、孕激素受体显著相关。COX 2与HER 2的表达密切相关 ,HER 2阳性乳腺癌中COX 2阳性表达率明显高于HER 2阴性者。结论 检测COX 2和HER 2表达与乳腺癌生物学行为有关 ,COX 2可能是HER 2导致肿瘤发生的机制之一

Abstract:Objective To study the expression of cyclooxygenase-2 (COX-2) and HER-2 in human breast cancer and their relationship. Methods Archival breast carcinoma (n=80), atypical hyperplasias (n=27) and normal breast epithelia (n=20) were analyzed for COX-2 and HER-2 expression by immunohistochemistry using specific monoclonal antibodies. Results COX-2 was expressed in 57.5% (46/80) of breast carcinomas at a moderate to high level, which was significantly different from the expression in benign breast epithelia. COX-2 expression was correlated with histological grade, lymph node metastasis, TNM stage and ER state (P< 0.05 or P< 0.01). HER-2 was overexpressed in 37.5% (30/80) of breast carcinomas. HER-2 expression level was correlated with TNM stage, lymph node metastasis, histological grade, ER state and PR state (P< 0.05 or P< 0.01). The expression of COX-2 in HER-2 positive breast cancers was significantly higher than that in HER-2 negative breast cancers. Conclusion Both COX-2 and HER-2 overexpression is significantly related to the biological behavior of human breast cancer. COX-2 is probably one of the most important mediators of HER-2 tumorigenic pathway.
