TGF-β_1中和抗体在预防肌腱黏连中的应用

目的探讨应用TGF-β1多克隆抗体抑制术后屈指肌腱黏连的效果。方法将108只来亨鸡随机分成3组,行中趾屈指肌腱切断吻合术,术中分别滴加生理盐水或不同浓度的TGF-β1多克隆抗体,于术后1、3、8周取肌腱行HE染色、Masson胶原染色、苦味酸-天狼星红染色和TGF-β1免疫组化染色。结果TGF-β1多克隆抗体10μg/mL组炎症过程缩短,塑形过程加速;TGF-β1多克隆抗体5μg/mL组、10μg/mL组Ⅰ胶原含量减少,且Ⅰ/Ⅲ胶原比例下降,TGF-β1的表达降低。结论TGF-β1多克隆抗体可以有效地抑制TGF-β1在肌腱损伤修复过程中的作用,减少肌腱的黏连、瘢痕组织的形成,这可能是治疗屈指肌腱损伤术后黏连潜在的有效方法之一。

The preventive effect of TGF-β1 neutralizing antibody on flexor tendon adhesion from operation

Objective To investigate the preventive effect of TGF-β1 neutralizing antibody on flexor tendon adhesion from operation.Methods Totally 108 leghon cocks performed anastomosis operation were divided into three groups randomly: normal saline(control group),5μg/mL group,and 10μg/mL group of TGF-β1 antibody.At week 1,3,and 8,respectively,after operation,HE staining,Masson staining,Sirius red-polarization staining and TGF-β1 immunohistochemistry staining were used.Results Compared with the control group,the 10μg/mL group had less shortened progress of inflammation and accelerated progress of molding;in the experimental groups(5μg/mL group,10μg/mL),the density of the collagen Ⅰ type was smaller,the ratio of Ⅰ/Ⅲ collagen and expression of TGF-β1 decreased.Conclusion TGF-β1 multiclonal neutralizing antibody can effectively inhibit the function of TGF-β1 during the flexor tendon repair,reduce tendon adhesion and scar formation,suggesting that it is a latent and effective method for preventing flexor tendon from adhering after operation.
