Objective To investigate the relationship between erythrocyte immune function and selenium (Se) level. Methods Forty-nine Kashin-Beck patients in endemic area aged 13- 1 6 years were divided into two groups and were orally given either selenized yeast or sodium selenite to provide 200 μg selenium per day for12 weeks. Erythrocyte selenium level, glutathione peroxidase activity, the rosette formation rates of red blood cells complement receptor typeⅠ (CR1), the immune function of red blood cells, and circulating immune complexes(CIC) were determined. Results After supplementing with selenium for 12 weeks, erythrocyte selenium level, glutathione peroxidase activity, the rosette formation rates of red blood cells CR1 were significantly increased. But the difference in rosette formation rates of IC and CIC content was not significant between before and after Se supplementation. Gonclusion The increase of the immune function of the erythrocyte by selenium-supplement may be one of the effective mechanisms for the prevention of Kashin-Beck disease.  相似文献

目的 探讨TNF-α基因在肺癌组织中的表达及其临床意义。方法 应用免疫组化SP法检测40例肺癌组织中肿瘤坏死因子(Tumor necrosis factor,TNF-α)的表达,并与癌旁正常肺组织对照。结果 TNF-α阳性表达主要分布于癌细胞胞浆中,阳性率为47..5%(19/40),细胞核内无阳性表达;癌旁正常肺组织内无阳性表达(P<0..05)。结论 TNF-α与肺癌的发生、发展有着密切关系。  相似文献

Objective To explore the roles of cytokines in the pathogenesis of hemorrhagic fever with renal syndrome(HFRS). Methods Double-antibody sandwich ELISA was used to determine serum interleukin (IL)-6, urine tumor necrosis factor (TNF), IL-6 and IL-8 levels in 56 patients with HFRS. Results Serum IL-6, urine TNF, IL-6 and IL-8 concentrations in HFRS patients were significantly higher than those in control group, respectively (P〈0. 001). The concentrations increased at fever stage, then continued to increase during hypotension stage and peaked at oliguria stage. The concentrations of serum IL-6, urine TNF, IL-6 and IL-8 increased in accord with the severity of the disease and differed greatly among different types of the disease. Serum IL-6 had remarkable relationships with serum specific antibodies. It was positively related to serum β2 -microglobulin (β2 -MG), blood ureanitrogen (BUN) and creatinine (Cr). Significant positive relationships were also found both between urine IL-6 and TNF, and between IL-6 and IL-8 (r=0.5768, P〈0.05; r=0. 3760, P〈0.01). Conclusion TNF, IL-6 and IL-8 activated during the course of the disease. IL-6 is associated with the immunopathological lesions caused by the hyperfunction of humoral immune response. IL-6, IL-8 and TNF are involved in the renal immune impairment. Determining them might, in certain extent, be used in predicting the prognosis and outcome of patients with HFRS.  相似文献

Objective To evaluate the impacts of rosiglitazone and metformin on ovarian response, hirsutism and insulin action in women with polycystic ovary syndrome (PCOS). Methods Ninety women resistant to clomiphene citrate with PCOS were randomized, 40 cases to rosiglitazone group, 50 cases to meformin group. Rosiglitazone and metformon were administered for 6 months in combination with clomiphene citrate on cycle day 5th to 9th, respectively. The clinical evaluations were performed monthly. Reproductive hormone, serum glucose and insulin levels were observed before and after treatment. The data were analyzed using repeated analysis of variance (ANOVA). Results There were significant changes in reducing hirsutism score, serum testosterone level, LH/FSH ratio, and restoration of ovarian ovulation after metformin or rosiglitazone administration respectively (P<0.05). Insulin sensitivity was more significantly improved in the rosiglitazone group than in metformin group (P<0.05) After treatment. Homa IR and Homa β were decreased from 1.54±0.34 to 0.83±0.38 and from 5.83±0.55 to 4.95±0.54 (P<0.05) in the rosiglitazone group. The body mass index(BMI) was decreased from 25.30±3.64 to 23.83±2.32 in the metformon group (P<0.05). There were no significant changes in ovulation rate and pregnancy rate between groups (P>0.05). Conclusion Rosiglitazone can increase insulin sensitivity. Metformon may reduce BMI. They all restore regular menstrual cycles, increase pregnancy rate, and reduce testosterone and LH concentration in women with polycystic ovary syndrome.  相似文献

Objective According to the distribution of low selenium areas, low nutrition state of the residents and the affecting cartilage growth and articular cartilage of Kashin-Beck Disease(KBD),the chondrocyte differentia- tion and differential expression of collagen types Ⅰ , Ⅱ and Ⅹ in articular cartilage from Chinese mini-pigs treated with low selenium were investigated in order to gain insight into the effects of these conditions on chondrocyte differ- entiation in KBD cartilage. Mothods Eleven male juvenile mini-pigs, aged from 4 weeks to 6 weeks after birth, were divided into 3 groups. The Se content in the diet of the “low Se” group was 0. 035mg/kg diet, and 0. 175 mg/kg diet in the control. For Se-supplemented group 0. 390mg /kg diet was added. The content of Se in blood was assayed at the beginning and at the end of each experiment. Samples of articular cartilage were taken from the right femur condylus, and collagen types Ⅰ , Ⅱ and Ⅹ in articular cartilage were analyzed by immunohistochemistry and in situ hybridization. Results ①All cartilage samples from juvenile mini-pigs fed with low selenium diet revealed a re- duction in type Ⅹ collagen mRNA expression in the hypertrophic chondrocytes as shown by in situ hybridization, and reduced type Ⅹ collagen deposition in the lower hypertrophic zone as shown by immunohistochemistry. ②Addition of selenium to the diet restored the type Ⅹ collagen to normal level. ③Type Ⅱ collagen was evenly distributed over the entire articular cartilage in all experimental and control groups. Type Ⅱ collagen mRNA signals were most prominent in the upper articular layer as well as in the hypertrophic zone in all groups. Type Ⅱ collagen expression was restrict- ed to the zone of endochondral ossification in all experimental groups and the control. Conclusion Low selenium has an down-regulatory role on the synthesis and deposition of collagen type Ⅹ in hypertrophic chondrocytes in articular cartilage of mini-pigs. Supplement of the low Se diet with additional Se restored the signals of collagen type Ⅹ to nor- mal levels. These findings indicate that selenium deficiency may disturb chondrocyte differentiation to hypertrophic cells in the growth plate,and worthy to be investigated further.  相似文献

目的 研究NO、TNF α、IL 1β、IL 6在突出腰椎间盘组织中的表达 ,并探讨其意义。 方法 用免疫组化方法 ,对2 0例腰椎间盘突出症患者的突出椎间盘组织iNOS、TNF α、IL 1β、IL 6的表达进行检测 ,并取 12例健康成人 (尸体 )正常椎间盘作为对照。结果  2 0例突出腰椎间盘组织中 ,iNOS、TNF α、IL 1β、IL 6表达阳性分别为 16、15、12、11例 ,其中 9例为四种细胞因子均表达阳性。 12例正常椎间盘组织中无表达阳性细胞。结论 突出腰椎间盘可产生上述细胞因子 ,阳性细胞主要以成纤维细胞、软骨细胞及淋巴细胞为主 ,这些细胞因子可能在椎间盘退变中发挥作用  相似文献

目的观察芪倍合剂对溃疡性结肠炎模型大鼠NF-κB活性、NF-κB mRNA及TNF-αmRNA表达的影响并探讨与抗脂质过氧化有关的作用机制。方法雄性SD大鼠75只,随机分为正常对照组、模型组和芪倍合剂小剂量治疗组[1.5 mL/(100 g.d)]、中剂量治疗组[2.0 mL/(100 g.d)]、大剂量治疗组[2.5 mL/(100 g.d)]。以三硝基苯磺酸(TNBS)/乙醇溶液灌肠诱导大鼠溃疡性结肠炎模型,治疗组造模同时给予芪倍合剂灌肠,共4周。造模4周后处死大鼠取结肠组织标本,光镜观察结肠组织的病理变化,并对结肠组织标本进行病理评分;免疫组化法测定结肠黏膜NF-κB活性,RT-PCR法检测结肠组织中NF-κB mRNA和TNF-αmRNA的表达,放射免疫法测定血清丙二醛(MDA)含量和超氧化物歧化酶(SOD)、GSH-Px的活性。结果与正常对照组比较,模型组结肠组织病理评分、MDA含量、NF-κB活性及TNF-αmRNA的表达显著增加(P<0.01),而血清SOD、GSH-Px的活性明显下降(P<0.01);与模型组比较,治疗各组病理评分、MDA含量、NF-κB的活性及TNF-αmRNA的表达显著下降,而血清SOD、GSH-Px的活性明显上升(P<0.05或P<0.01)。NF-κB的活性及TNF-αmRNA的表达呈正相关关系(r=0.570,P<0.05),各组NF-κB mRNA的表达均为阳性,无显著性差异(P>0.05)。结论芪倍合剂能显著降低实验性溃疡性结肠炎大鼠的NF-κB的活性、TNF-αmRNA的表达,从而减轻结肠组织损害程度,这与其抑制抗脂质过氧化作用密切相关。  相似文献
