目的构建含有人肠三叶因子基因cDNA的腺病毒载体,并观察其对肠上皮细胞的感染能力。方法利用PCR技术扩增hITF基因cDNA全长序列,测序后亚克隆至腺病毒穿梭质粒,转化至含腺病毒骨架质粒pAdEasy-1大肠杆菌株BJ5183感受态细胞中进行同源重组,内切酶PmeⅠ线性化后转染HEK293细胞进行病毒包装及扩增,并进行滴度测定。将重组腺病毒感染肠上皮细胞(HT-29),荧光显微镜观察感染效果。结果成功构建出重组穿梭质粒pAdTrack-CMV-hITF,PmeⅠ线性化后与骨架质粒pAdEasy-1同源重组,PacⅠ酶切鉴定后命名为Ad-hITF;重组腺病毒载体在HEK293细胞中包装,获得重组腺病毒颗粒;根据Karbers公式计算病毒颗粒滴度为2×109 pfu/mL;重组腺病毒可以感染肠上皮细胞(HT-29),感染效率较高。结论成功构建含hITF基因cDNA的重组腺病毒载体并获得大量子代重组腺病毒,为腺病毒介导hITF基因治疗的应用打下基础。 相似文献

Abstract A watershed‐based approach to water quality management is necessary for protecting coastal water quality, but the relative importance of inland pollution sources on estuaries is often overestimated. This can lead to a misallocation of coastal water protection resources and ignorance of coastal zone pollution sources. This overestimation of upstream contributions is attributable to a failure to recognize that many estuaries have little or no inland drainage area, the confusion of basinwide pollutant loading with pollutant delivery to estuaries, the low delivery ratios for many pollutants within drainage basins, and disproportionately high pollutant delivery for sources within the coastal zone. Examples are given from North Carolina. As a general rule, resources expended on pollution control within or near the coastal zone will result in more coastal water quality improvement per unit effort than resources expended upstream. 相似文献

ABSTRACT As maintenance and operation costs increase with usage over time, equipment is replaced when the value of new equipment is more attractive. Some methods have been developed to solve this problem. In the public transport sector, such problems are frequently analyzed by fleet managers and determined by bus age restriction regulations. We propose an Integer Programming model that integrates both budgetary and environmental constraints (CO2 emissions) which, as far as we know, have not previously been studied in conjunction. The study aims to determine the optimal replacement plan for a fleet of diesel buses of different size, age, maintenance costs and emissions rates, with new (less polluting) diesel buses over a time horizon of 50 years. The results indicate that it is possible to reduce emissions with a low annual budget using an optimal replacement policy. 相似文献

A trio of closely related land‐use/transport interaction models was developed using Marcial Echenique & Partners’ software package MEPLAN. The models were for the cities of Bilbao, (Spain), Dortmund (West Germany) and Leeds (England). All were calibrated using data drawn from earlier studies. The three models were used to predict the effects (relative to a base case) of a common programme of land‐use and transport policies or scenarios. In this article, the author examines the concept of piracy under international law and related notions such as hijacking and mutiny. ABSTRACTGovernments require decision tools to deal with road traffic accidents, a pandemic resulting in millions of deaths around the world. Evidence shows that human factors are one of the major causes of road accidents, and there is much interest in identifying variables that may have an impact on drivers’ perception of risk. To this aim, we design a stated choice experiment with eight hypothetical driving scenarios considering attributes that have been strongly associated with increased accident risks: (i) driving speed, (ii) driving the wrong way in a one-way street, (iii) overtaking on a bend, and (iv) driving under the influence of alcohol or drugs. Data from a sample of survey respondents are used to estimate a hybrid discrete choice model incorporating two latent variables, Driver Concentration and Safe Driving. Our results may contribute to the design of public policies geared to prevent accidents by encouraging safer driving behaviour. 相似文献

Research to quantify the potential benefits and drawbacks of practice, and to identify barriers to adoption, is the first step in any strategic plan for their adoption. However, uncertainties, complexity, interdependence, and the multidisciplinary nature of emerging transportation technologies make it difficult to organize and identify focused research. The contribution of this work is a cognitive framework to help planners and policymakers organize broad topics, reveal challenges, discover ideas for solutions, quantify potential impacts, and identify implications to guide preparation strategies. [Objectives ] This paper studies a three-dimensional (3D) cooperative path-following control problem in the process of maritime search and rescue for a heterogeneous unmanned cluster system composed of unmanned aerial vehicles (UAVs) and unmanned surface vehicles (USVs).[Methods ] First, kinematic models of the UAVs and USVs are established under a fixed coordinate system and body coordinate system. In order to design a 3D path-following controller suitable for motion control, an air coordinate system is established, and the path tracking error models of the UAVs and USVs are established in the Serret-Frenet coordinate system. Next, a 3D line-of-sight (LOS) guidance law is designed at the kinematic level, and a cooperative path-following control method suitable for heterogeneous clusters of marine vehicles is proposed, allowing the UAVs and USVs to track the preset parameterized path. Finally, the stability of the control system is analyzed based on the Lyapunov stability theory.[Results]The simulation results verify the effectiveness of the proposed cooperative path-following control method for heterogeneous clusters of marine vehicles.[Conclusions]The results of this study can provide references for maritime search and rescue by using the proposed cooperative path-following control method. © 2022 Journal of Clinical Hepatology.
