SIMPLE METHOD OF PUNCHING AND RE-LOCATING TISSUES FOR MANUAL CONSTRUCTION OF TISSUE MICROARRAY

A series of human tissue samples and cultured cell lines were formalin-fixed and paraffin-embedded. Specimen cylinder ( 1.2 - 1.8mm) were punched by a modified bone marrow biopsy needle and arrayed on a recipient paraffin block. Microscopic analysis on the sections from this tissue microarray (TMA) block demonstrated that the spots of tissues and cells were well preserved, and the cultured cell samples were successfully embedded from 5 × 104 to 2 × 105 in number. These TMA sections were also suitable for immunohistochemistry and RNA in situ hybridization.     

LIGUSTRAZINE REDUCES CARTILAGE DESTRUCTION IN MODEL OF OSTEOARTHRITIS IN RATS

Objective To study the effect and possible mechanism of Ligustrazine-a chinese traditional herbal medicine (CTM) on rat osteoarthritis (OA). Methods OA model was surgically induced. Morphology of articular cartilage was done by HE staining and mankin score was calculated; interleukin-1 and TNFα activity of rat joints was determined by 3-(4,5 dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) method; immunohistochemistry of MMP-13 and Cathepsin K was done by ABC method while the mRNA level for MMP-13, cathepsin K and tissue inhibitor of matrix metalloproteinase 1 (TIMP-1) was evaluated by RT-PCR method. ResultsLigustrazine (60 mg·kg-1·d-1 or 30 mg·kg-1·d-1, but not 10 mg·kg-1·d-1, im, ×8 weeks) reduced morphological changes of articular cartilage of OA rats. IL-1 and TNFα production of OA joints increases, together with MMP-13, cathepsine K and their mRNA levels in articular cartilage. The administration of Ligustrazine (60 mg·kg-1·d-1or 30 mg·kg-1·d-1, im, ×8 weeks) reduced above changes significantly. At the same time, tissue inhibitor of metrix metalloprotease (TIMP1) mRNA level increased also in OA rats while Ligustrazine (60 mg·kg-1·d-1, 30 mg·kg-1·d-1or 10 mg·kg-1·d-1, im, ×8 weeks) had no significant effect on it. Conclusion Ligustrazine (60 mg.kg-1 d-1 or 30 mg·kg-1·d-1, but not 10 mg·kg-1·d-1, im, ×8 weeks) effectively delays articular cartilage degradation of OA rats and could be a potential drug candidate for OA treatment.     

INTRAOPERATIVE LOCALIZATION OF CORTICAL MOTOR EVOKED POTENTIALS IN CENTRAL SULCUS LESIONS

Objective To study direct cortical electrical stimulation technique for the recording of motor evoked potentials under general anesthesia in central sulcus lesions. Methods The largest N20-P25 response was recorded from postcentral gyrus by intraoperative monitoring of cortical motor evoked potentials in 10 patients with intracranial lesions near or in the central area. The muscles of upper extremity in all patients were activated by delivering stimulus to cortical areas continuously. Moving the cortical electrodes forward, the largest P20-N25 response, SEP phase reversal,was obtained as a motor center stimulus. In this site of cortex, a short train stimulation elicited reproducible muscle action potentials that could be observed from the oscilloscope without averaging.Results MEPs can be recorded, pre- and post-operatively, without motor deficits of upper limbs in all patients.Conclusion This technique seems to be preferable for intraoperative localization of motor evoked potentials in central sulcus lesions under total intravenous anesthesia.

Effects of Dynamic Changes in Ultrasound Attenuation and Blood Perfusion on Lesion Formation of Multiple focus Pattern during Ultrasound Surgery

A nonlinear finite-element program was developed to simulate the dynamic evolution of coagulation in tissue considering temperature and thermal-dose dependence of the ultrasound attenuation and blood perfusion rate.The effects of these dynamic parameters on the lesion formation were investigated in the particular case of ultrasound hepatic ablation with bi-focus intensity pattern.The results of simulations were compared that incorporate dynamic changes of ultrasound attenuation and perfusion and results that neglect these effects.The result shows that thermal-dose-dependent ultrasound attenuation is the dominating factor in the full dynamic model.If the dynamic ultrasound attenuation is ignored, a relatively significant underestimation of the temperature rise appears in the focal plane and the region next to the focal plane, resulting in an underestimation in predicting diameter of coagulation.Higher heating intensity leads to greater underestimation.     

Secure Authentication of WLAN Based on Elliptic Curve Cryptosystem

The security of wireless local area network (WLAN) becomes a bottleneck for its further applications. At present, many standard organizations and manufacturers of WLAN try to solve this problem. However, owing to the serious secure leak in IEEE802.11 standards, it is impossible to utterly solve the problem by simply adding some remedies. Based on the analysis on the security mechanism of WLAN and the latest techniques of WLAN security, a solution to WLAN security was presented. The solution makes preparation for the further combination of WLAN and Internet.     

Investigation of Wall Pressure Fluctuations in a Turbulent Boundary Layer by Large Eddy Simulation

Large eddy simulation (LES) was used to investigate the space-time field of the low Mach number, fully developed turbulent boundary layer on a smooth, rigid flat plate. The wall-pressure field simulated by LES was analyzed to obtain the pressure statistics, including the wall-pressure root-mean square, skewness and flatness factors, which show the wall pressure distribution was not Gaussian. The profile of the auto-power spectral density and the contour of the streamwise wavenumber-frequency spectral density of wall-pressure were plotted. The "convection ridge" can be observed clearly and the convection velocity can be calculated from the location of the convection peak.
