VEGF-C、VEGFR-3和podoplanin在食管癌中的表达及其意义

目的观察血管内皮生长因子C(VEGF-C)及其受体3(VEGFR-3)和肾小球上皮细胞整合膜蛋白(podoplanin)在食管癌组织中的表达,分析其与肿瘤淋巴结转移的关系。方法免疫组织化学SP法检测76例食管癌组织中VEGF-C、VEGFR-3和podoplanin的表达及淋巴管密度(LVD)。结果食管癌VEGF-C表达阳性率为63.1%,VEGF-C的表达与食管癌的淋巴结转移、TNM分期有统计学意义(P<0.01,P<0.05),但与患者年龄、肿瘤大小及分化程度无关;癌旁组织淋巴管密度同淋巴结转移、VEGF-C阳性表达间有统计学意义(P<0.01,P<0.05),podo-planin LVD值与VEGFR-3 LVD值相比,具有统计学意义(P<0.01)。结论食管癌组织中VEGF-C表达与肿瘤TNM分期、淋巴管密度和淋巴结转移有关,VEGF-C表达越高,淋巴管密度越大,淋巴结转移可能性越高。癌旁组织LVD同VEGF-C的高表达及淋巴结转移密切相关,食管癌组织中podoplanin标记的淋巴管密度较VEGFR-3标记的淋巴管密度更为精确,是一种更为满意的淋巴管内皮特异性标志物。     

氧化苦参碱对慢性肾脏病患者血清TGF-β1和Col Ⅲ的影响

目的观察氧化苦参碱对慢性肾脏病(CKD)患者血清转化生长因子-β1(TGF-β1)、Ⅲ型胶原(ColⅢ)的影响,探讨其在肾间质纤维化发生发展过程中的作用和治疗价值。方法选取健康对照者10例、慢性肾脏病患者40例,根据肾小球滤过率(GFR)分为轻度肾损害组(GFR<90 mL/min.1.73 m2,CKDⅠ)、中重度肾损害组(GFR<60 mL/min.1.73 m2,CKDⅡ),再分层随机抽样,分为常规治疗组和氧化苦参碱组。利用双抗体夹心酶联免疫吸附法分别检测各研究对象治疗前、后血清TGF-β1、ColⅢ含量,采用t检验比较两组TGF-β1、ColⅢ水平的差异。结果氧化苦参碱组TGF-β1、ColⅢ含量显著低于常规治疗组(P<0.05)。结论氧化苦参碱可能通过下调TGF-β1、ColⅢ水平,减少ECM沉积,从而达到防止肾间质纤维化的作用。     

The role of interleukin-8 produced by tumor induced fibroblasts in the development of cutaneous melanoma

Objective To determine the role of interleukin-8 (IL-8) produced by tumor induced fibroblasts in the development of cutaneous melanoma. Methods B16 melanoma cells induced L929 fibroblasts phenotype was transdifferentiated to myofibroblasts (MF) by co-culture in vitro. MF was monitored by morphology and immunophenotype for a-SMA. The level of IL-8 was detected by ELISA. The effect on B16 cell proliferation rate was estimated using MIT method in vitro. Melanoma implanting model was constructed in C57 mice. Results L929 MF phenotype could be modulated by B16 melanoma cells-derived transforming growth factor-β1 (TGF-β1) and elevated the levels of IL-8. L929 MF did not influence the B16 melanoma cells viability in vitro, but shortened the time of tumor formation and increased the incidence rates of tumors in C57 implanting model mice. Conclusion Fibroblasts can be activated by tumor cells and produce IL-8, which acts as an inflammatory cytokine promoting the development of cutaneous melanoma.

Wind pressure distribution and wind-induced dynamic response for spatial groined latticed vaults

The wind pressure distribution and wind-induced vibration responses of long-span spatial groined latticed vaults (SGLVs) were numerically simulated, which always are ones of the most important problems in the structural wind resistance design. Incompressible visco-fluid model was introduced, and the standard k-εtwo equation model and semi-implicit method for pressure linked equation (SIMPLE) were used to describe the flow turbulence. Furthermore, the structural dynamic equation was set up, which is solved by Newmark-β method. And several sort of wind-induced vibration coefficients such as the wind-induced vibration coefficient corresponding to the nodal displacement responses and wind loads were suggested. In the numerical simulation where the SGLV consisting of the cylindrical sectors with different curved surface was chosen as the example,the influence on the relative wind pressure distribution and structural wind-induced vibration responses of the closed or open SGLV caused by such parameters as the number of cylindrical sectors, structural curvature and the ratio of rise to span was investigated. Finally, some useful conclusions on the local wind pressure distribution on the structural surface and the wind-induced vibration coefficients of SGLV were developed.
