THE EXPRESSION OF BCL-2, BAX AND CASPASE-3 IN NEURON OF THE SPINAL CORD ANTERIOR HORN AFTER CAUDA EQUINA ACUTE COMPRESSION

Objective To explore the influence of the acute cauda equina compression on the iumbosacral spinal cord; To clarify the pathologic change of the motor neuron after acute cauda equina compression. Methods 27 canis familiaris were randomly divided into 9 groups （3 in each） ： one for normal group, one for control group, and seven for compression groups. The control group and compressed groups was given operation and the sac made of silica gel was placed under the lamina of L5-6. Water was injected into the sac until their posterior legs paralysis in compressed groups, the animals had been compressed for 4, 8, 12, 24, 48, 72, 168 hours. The control group were not injected water. Cells apoptosis was investigated with the technology of TdT-mediated biotin dUTP nick end-labeling （TUNEL） staining. The Bcl-2 Bax and Caspase-3 protein was investigated by immunohistochemical method. Results TUNEL staining cells in anterior horn presented after compressed 8-12 hours, and at 72 hours the number of positive cells got to maximum, it decreased subsequently after 168 hours. The protein of Bax, Bcl-2 expressed a little in normal motor neuron. The caspase-3 protein didn＇t express in normal ceil. They all reached the peak at 72 hours after compression. Conclusion The apoptosis of motor neuron occurred earlier after eauda equina acute compression. Bax protein restrained Bcl-2 protein then active caspase-3 and conduced apoptosis of motor neuron.     

罗红霉素胶囊在人体的药代动力学及生物等效性

目的建立人血浆中罗红霉素浓度的高效液相分析方法,用于研究罗红霉素胶囊在人体的药代动力学和生物等效性。方法血浆样品采用己烷-异戊醇(98∶2)液-液萃取,反相高效液相法测定。结果罗红霉素的线性范围为0.25-32.06 mg/L(r为0.999 2),最低检出质量浓度为0.25 mg/L,最低检出量为12.5 ng,提取回收率为68.0%-72.8%,方法回收率为90.7%-99.4%。单次服用300 mg罗红霉素胶囊受试制剂或参比制剂后的药动学参数AUC0-48h、Cmaxt、maxt、1/2分别为(101.84±27.69)(mg.h)/L和(103.5±30.83)(mg.h)/L,(8.54±1.95)mg/L和(8.07±1.81)mg/L,(1.6±0.6)h和(1.8±0.6)h,(10.4±2.9)h和(10.5±2.6)h。受试制剂相对生物利用度为(100.5±19.1)%。结论该方法专一性较好,血浆内源性杂质无干扰,结果准确。罗红霉素的两种制剂间主要药动学参数无明显差异,具有生物等效性。     

功能性矫治器引导大鼠下颌前伸后咀嚼肌基质金属蛋白酶mRNA的表达

目的研究功能性矫治器引导大鼠下颌前伸后咀嚼肌中基质金属蛋白酶(MMPs)mRNA的表达,探讨功能性颌骨矫形过程中咀嚼肌适应性变化的生物学机制,为矫形治疗提供新的理论依据。方法分别采用RT-PCR法、实时荧光定量PCR方法观察戴矫治器(实验组)和不戴矫治器(对照组)3、7、14、21 d大鼠二腹肌前腹中MMP-2、MMP-9mRNA表达和表达差异。结果①实验组和对照组大鼠二腹肌前腹中MMP-2、MMP-9 mRNA均有表达。②MMP-2mRNA、MMP-9 mRNA表达差异:实验组3、21 d后表达增加,其中21 d较14、7 d表达明显增加;而对照组21 d较143、d表达明显增加。结论①MMP-2、MMP-9在mRNA水平参与了咀嚼肌正常的生长发育过程;②功能性矫治器引导大鼠下颌前伸后咀嚼肌中MMPs在mRNA水平参与了咀嚼肌的适应性变化。     

Mirror Mapping of Laser Interferometer Measurement System on Ultra-precise Movement Stage

The accuracy and repeatability of the laser interferometer measurement system （LIMS） are often limited by the mirror surface error that comes from the mirror surface shape and distortion. This paper describes a new method to calibrate mirror map on ultraprecise movement stage （UPMS） with nanopositioning and to make a real-time compensation for the mirror surface error by using mirror map data tables with the software algorithm. Based on the mirror map test model, the factors affecting mirror map are analyzed through geometric method on the UPMS with six digrees of freedom. Dam processing methods including spline interpolation and spline offsets are used to process the raw sampling data to build mirror map tables. The linear interpolation as compensation method to make a real-time correction on the stage mirror unflatness is adopted and the correction formulas are illuminated. In this way, the measurement accuracy of the system is obviously improved from 40 nm to 5 nm.

男性不育机制部分研究热点的探讨

半个世纪以来,男性不育患者的比例有升高趋势,但其病因和发病机制仍未完全阐明。本文结合我们的部分研究成果,概括介绍男性不育机制的部分研究热点,着重介绍精索静脉曲张、甲醛及男性不育相关基因与男性不育的可能关系。     

Tenascin、CD34在胆管癌组织中的表达及临床意义

目的探讨Tenascin、CD34在胆管癌的发生、发展中的作用和意义。方法收集我院1995年-2003年期间切除的胆管癌标本33例和正常胆管组织4例,采用免疫组织化学S-P方法,检测其Tenascin、CD34的表达情况。结果Tenascin在胆管癌组织中的阳性表达率为81.8%(27/33),其中病理分级Ⅰ级、Ⅱ级、Ⅲ级的阳性率分别为71.4%、82.4%和88.9%,但无显著性统计学差异(P>0.05);在正常胆管组织中未检测到Tenascin表达。CD34在胆管癌组织中广泛表达,在正常胆管组织中仅有少量表达,两者相比具有显著统计学差异(P<0.05)。结论Tena-scin可以作为胆管癌的肿瘤标志应用于临床诊断,CD34可作为预测肿瘤转移的一项重要指标。     

镓盐对骨质疏松骨细胞超微结构的影响

目的探讨镓盐对骨质疏松骨细胞超微结构的影响。方法用8月龄雌性SD大鼠造模,骨质疏松组开腹切除双侧卵巢后缝合,对照组开腹切一小块脂肪后缝合。12周后,取胫骨上段骨组织,观察模型复制情况。嗣后随机分为3组:对照组与骨质疏松组正常饲养12周;硝酸镓治疗组给予硝酸镓治疗(1 mg/kg),3次/周,腹腔注射。应用脱钙骨切片观察光镜下骨形态;制作透射电镜样本,在透射电镜下观察骨细胞形态及骨细胞内部结构情况。结果对照组及硝酸镓治疗组骨细胞结构正常,细胞核染色质分布均匀,核为卵圆形;骨质疏松组部分骨细胞核呈固缩状,形态变长。对照组成骨细胞中细胞器较少,有胶原分泌;骨质疏松组成骨细胞中有大量内质网存在,有大量胶原分泌,硝酸镓治疗组成骨细胞可见有较多胶原分泌,细胞中存在大量内质网及线粒体。结论镓盐对骨质疏松引起的骨细胞超微结构的损伤有改善作用。     

薄壁铜及铜合金高频焊焊缝接头的错位研究

分析了薄壁铜及铜合金高频焊接焊缝接头的错位及其产生的主要原因,研究了焊缝接头错位与焊接过程稳定性及焊缝强度的关系,提出了焊缝接头错位的预防和控制方法。
