ROLE OF PERIPHERAL LYMPHOCYTES SIALYL LEWIS（X） （CD15s） ANTIGEN BEFORE AND AFTER KIDNEY TRANSPLANTATION

Objective To investigate the role of peripheral lymphocytes Sialyl Lewis（x） （CD15s） antigen before and after kidney transplantation. Methods Flow cytometry technique was applied to examine the expression of peripheral lymphoid cell surface CD15s antigen after renal transplantation, and to evaluate various therapeutic regimen. Results The statistic analysis results of peripheral lymphoid cell surface CD15s antigen expression level showed that there was significant difference among the patients with acute rejection, long-term dialysis and with normal renal function post-transplant; significant difference of CD15s expression level between group of rejection and infection; no significant difference of CD15s expression among the different groups treated by various therapeutic regimens. Conclusion The different therapeutic regimen has no influence to CD15s expression; Detection of peripheral lymphoid cell surface CD15s antigen expression periodically, intelligently make convenience to understand suitable status of immunosuppression.     

CLASSIFICATIONS OF EEG SIGNALS FOR MENTAL TASKS USING ADAPTIVE RBF NETWORK

Objective This paper presents classifications of mental tasks based on EEG signals using an adaptive Radial Basis Function (RBF) network with optimal centers and widths for the Brain-Computer Interface (BCI) schemes. Methods Initial centers and widths of the network are selected by a cluster estimation method based on the distribution of the training set. Using a conjugate gradient descent method, they are optimized during training phase according to a regularized error function considering the influence of their changes to output values. Results The optimizing process improves the performance of RBF network, and its best cognition rate of three task pairs over four subjects achieves 87.0%. Moreover, this network runs fast due to the fewer hidden layer neurons. Conclusion The adaptive RBF network with optimal centers and widths has high recognition rate and runs fast. It may be a promising classifier for on-line BCI scheme.     

离体毛囊切片两种制作方法的比较及凋亡检测

目的对人头皮游离毛囊(HF)切片制作及凋亡检测方法进行比较。方法分别将石蜡切片法和冰冻切片法应用于游离毛囊;利用原位缺口末端标记法(TUNEL)进行荧光染色与3,3′-二氨基联苯胺(DAB)染色,检测未经培养的游离毛囊及培养5 d的游离毛囊的不同部位凋亡细胞情况。结果在对游离毛囊形态检测中,各组毛囊在外根鞘(ORS),毛球(HB)下部,真皮鞘(DS)及毛乳头(DP)中的凋亡细胞数并无显著性差异(P>0.05)。结论冰冻切片法应用于游离毛囊简单有效,TUNEL原位荧光结合DAB染色适合于检测毛囊细胞凋亡情况。

REAL-TIME DETECTION OF SURVIVIN mRNA EXPRESSION IN CERVICAL CANCER CELL LINES USING MOLECULAR BEACON IMAGING

Objective To detect the expression of survivin mRNA in cervical cancer cell lines using molecular beacon imaging technology. Methods Human cervical cancer cells （HeLa and SiHa） and human fetal lung fibroblast HFL-I were cultured in vitro. After adding 100 nmol/L survivin mRNA molecular beacon, the fluorescent signals were observed under fluorescent microscope. The expressions of survivin in cervical cancer cells and HFL-I cell were examined by immunocytochemical streptravidin-biothin peroxidase （SP） assay at the same time. Results Two kinds of survivin mRNA molecular beacon, with different color fluorescence, had strong fluorescent signal in cervical cancer cell lines, and the signal in SiHa cell line was stronger, but these signals were not found in HFL-I ; Immunocytochemical staining of positive survivin was located in the cytoplasm of cervical cancer cell lines HeLa and SiHa, whereas, no expression of survivin was detected in HFL-I cell line. Conclusion The technology of molecular beacon imaging can be used to detect the expression of survivin mRNA in viable cells successfully, and may provide a new approach to the diagnosis of early stage cervical cancer and the following-up in the clinic.     

硬膜外麻醉下持续靶控输注丙泊酚的镇静效果

目的　评价丙泊酚靶控输注 (TCI)在硬膜外阻滞麻醉不同镇静分级时的脑电双频谱指数 (BIS)与生命体征变化。方法　择期硬膜外麻醉下行中下腹部手术的 5 3例患者 ,采用自身对照 ,比较在不同Ramsay分级时 ,丙泊酚靶控输注的效应部位浓度 (ESC)与BIS值生命体征变化。结果　在Ramsay 4级时的ESC为 (1.6 6± 0 .16 )mg·L-1,BIS为 78.5± 4 .6 ,生命体征平稳 ,其HR、MAP、RRS、SPO2 与Ramsay 2级比较无差异 (P >0 .0 5 ) ;与Ramsay 5级、6级比较差异有显著性 (P <0 .0 1)。结论　随着丙泊酚靶控输注的效应部位浓度的增加 ,BIS值呈计量相关性下降并能指导镇静分级。在硬膜外阻滞麻醉下Ramsay镇静 4级时较为适宜 ,生命体征平稳 ,并发症少

全脑缺血再灌注损伤后JUN蛋白在大鼠海马的表达及热休克预处理对其表达的影响

目的研究全脑缺血再灌注损伤后JUN蛋白在大鼠海马的表达及热休克预处理对其表达的影响。方法以四血管法建立全脑缺血再灌注模型。将大鼠置于42℃中15 min行热休克预处理,全脑缺血6 min后行2 h、6 h、12 h、24 h3、d、5 d再灌注后处死,取海马脑组织行HE染色、JUN蛋白免疫组化染色,TUNEL法检测凋亡细胞。结果缺血再灌注后2 h JUN蛋白在CA1区开始表达,6 h达到高峰,5 d时仍有表达;CA3区JUN蛋白的表达弱于CA1区(P<0.05);同时CA1区细胞损伤明显。热休克预处理组JUN蛋白表达在CA1和CA3区相应时点减弱(P<0.05),细胞损伤轻微,凋亡细胞减少(P<0.05)。结论全脑缺血再灌注损伤后JUN蛋白过度表达参与了神经元损伤过程,热休克预处理通过下调JUN蛋白过度表达具有脑保护作用。     

海堤开口中海床防护设计探讨

结合厦门集杏海堤开口改造工程,对海床及临近桥墩等建构筑物受到的冲刷进行研究,并提出工程防护措施。同时,对国内外不同的冲刷深度计算公式进行分析、对比,为类似桥墩冲刷、海床防护工程提供参考。
