Fine Mapping of a Deafness Mutation hml on Mouse Chromosome 10

Objective To map a mouse deafness gene.identify the underlying mutation and develop a mouse model for human deafncss.Methods Genetic lindage cross and genome scan wer used to map a novel mutation named hypoplasia of the membranous labyrinth (hml),which causes hearing loss in mutant mice.Results ① hml was mapped on mouse Chr 10 (～43 cM from the centromere) suggests that the homologous human gene is on 12q22--q24, which was defined on the basis of known mouse-human homologies (OMIM,2004). ② This study has generated 25 polymorphic microsateUite markers, placed 3 known human genes in the correct order in a high-resolution mouse map and narrowed the hml candidate gene region to a 500 kb area.

目的　定位小鼠致聋基因 ,识别决定其性状的有关突变 ,为人类耳聋基因研究提供动物模型。方法　利用全基因组扫描来定位名为hml可致小鼠听力丧失突变基因。结果　①hml基因定位在小鼠 10号染色体上 ,距中心粒约4 3cM处。根据已知的鼠 人同源同线性特点 ,提示人的同源基因位于 12 q2 2 -q2 4 ;②获得了 2 5个多态性微卫星标记 ,通过高分辨的小鼠图谱将 3个已知人类基因进行了正确排列 ,并将hml侯选基因限定在一个 5 0 0kb的区域内。     

Train boarding and alighting rates at high passenger loads

Line capacity in metro and high‐frequency suburban railways is as much determined by station stop times as by factors such as line speed or train acceleration. This paper applies the method developed by London Underground to estimate the time that trains spend at stations, as a function of the physical characteristics of the situation (e.g. train door width) and the numbers of passengers involved. Analysis was carried out on a number of alternative designs for refurbishment of South West Trains' Class 455 inner‐suburban rolling stock. Whilst there is indeed an interaction between boarding and alighting passengers, this paper demonstrates that the LUL relationship breaks down at the highest passenger loads. Moreover, results indicate that passenger flow is not equal between different parts of the same group of boarders or alighters.

Using risk analysis to determine the potential range of lifecycle costs for urban transit systems

Intense competition for limited public funding for urban transport projects can often result in proponents of individual schemes presenting minimized costs and maximized benefits to funding bodies to try to ensure that their scheme is chosen above others for funding. This presents public bodies with a problem, especially in an era when they are keen to attract private contributions for transit schemes. Risk modelling techniques can be of great assistance in ascertaining ranges of costs and benefits for individual submissions and deciding upon which projects should receive priority-not only those with greatest cost-benefit indices, but also those whose indices have low levels of associated risk to allay the fears of the traditionally risk averse private sector.     

A study of rail demand in a two‐tier fares environment

A method based on logit analysis is suggested for predicting the effects on patronage and revenue of relative changes in peak and offpeak fares. The method also permits the ready estimation of consequent changes in service profitability. Data was collected specifically for this study from British Rail's Teesside corridor to achieve model calibration. Results show the extent to which demand in peak and offpeak fare‐periods is complementary when fare restrictions are applied only to the morning peak.
