Study of the mechanism on the apoptosis induced in Human leukemia cell line K562 by the combination of indole-3-acetic acid and horseradish peroxidase

Objective To investigate the mechanisms of apoptosis induced in Human leukemia cell line K562 by the combination of indole-3-acetic acid and horseradish peroxidase. Methods Human leukemia cell line K562 were exposed to indole-3-acetic acid （IAA） at 20, 40, 60, 80 or 100 mol/L and horseradish peroxidase（HRP） at 1.2 g/mL for varying times. MTT assay was applied to detect the cell proliferation. Flow cytometry was performed to detect the arrest of cell cycle. Terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling （TUNEL） assay was used to measure apoptosis. 2, 7-dichlorofluorescin diacetate （DCFH-DA） uptake was measured to determine free radical by confocal microscope. Content of malondiadehyde （MDA） and activity of superoxide dismutase （SOD） were measured by biochemical methods. Results IAA/HRP initiated growth inhibition of K562 cells in a dose- and time-dependent manner. Flow cytometry revealed that cell cycle arrested at G1/G0 after 24 hours treatment. After 72 hours treatment, apoptotic rate of 100 mol/L IAA group increased to 43.9 %, which was 5 times that of control（P〈0.01）. Content of MDA and activity of SOD increased respectively in treatments compared with control. Meanwhile, IAA/HRP stimulated the formation of free radical, which was increased by IAA concentration-dependently. Conclusion The combination of IAAand HRP can inhibit the growth of Human leukemia cell line K562 in vitro by inducing apoptosis which is associated with the increase of free radical. The combination of IAA and HRP might be a promising chemopreventive and chemotherapeutic agent against human leukemia.

IAA与HRP联合作用对人胰腺癌BXPC-3的细胞毒效应

目的　研究吲哚乙酸(IAA)与辣根过氧化物酶(HRP)在体外共同作用于人胰腺癌BXPC 3 细胞所产生的细胞毒效应。方法　MTT法测定IAA/HRP对BXPC 3细胞增殖的影响;流式细胞仪分析BXPC 3 细胞周期时相变化;胎盘兰(曲利本兰)排斥试验,研究IAA/HRP对BXPC 3 细胞死亡率的影响;原位细胞凋亡检测试剂盒检测IAA/HRP作用后BXPC 3细胞的凋亡。结果　IAA/HRP体外对BXPC 3 细胞有抑制生长的效应,且这种效应有浓度和时间依赖性;BXPC 3细胞在IAA/HRP作用48 h后细胞周期被阻滞在G1期和G2期;IAA/HRP作用72 h后对照组和药物组的凋亡细胞数存在着显著性差异,且凋亡细胞数有浓度依赖性。结论　IAA/HRP对胰腺癌BXPC 3 细胞有细胞毒作用,可体外抑制细胞的增殖,诱导凋亡。     

THE STUDY ON THE CHANGES OF ZINC, COPPER, CALCIUM AND MAGNESIUM IN PLASMA AND ERYTHROCYTES DURING CARDIOPULMONARY BYPASS

Objective To study the changes and their influence factors involved of zinc, copper, calcium and magnesium in plasma and erythrocytes during cardiopulmonary hypass(CPB). Methods Zinc, copper, calcium and magnesium values in plasma and erythrocytes were measured by atomic absorption spectrophotometer during CPB. Results Zinc and copper levels in plasma were significantly elevated above preinduction level before perfusion, hut calcium and magnesium levels did not change significantly; zinc, copper and calcium levels in plasma were significantly below preoperation level during CPB, hut magnesium level in plasma was significantly increased above preoperation; zinc level in plasma was increased to preoperation level after CPB and began to decrease again at 8 hours after CPB,copper level in plasma was increased to preoperation level at 20 hours after CPB, calcium in plasma was increased significantly from beginning to 8 hours after CPB, magnesium level in plasma was decreased to preoperation level at 8 hours afterCPB. Concentration of zinc , copper, calcium and magnesium in erythrocytes did not change significantly. Conclusion During CPB, the changes of zinc, copper, calcium and magnesium had relation to hemodilution, operative wound, carrier protein, stress and component of priming Solution and cardioplegic solution, hut no relation to transferfrom plasma erythrocytes. The results indicate that it is beneficial to patient‘s recovery to supplement zinc, copper, calcium and magnesium properly by different ways during cardiac perioperation.     

微波辐射下合成2-氨基-5-芳氧亚甲基-1, 3, 4-噻二唑

在三氯氧磷作用下,由芳氧基乙酸和氨基硫脲经脱水环合,微波辐射快速合成了一系列2-氨基-5-芳氧亚甲基-1, 3, 4-噻二唑类化合物.目标产物的结构经红外光谱及熔点测定得到确证.该方法具有反应时间短、操作简便、副产物少、产率高、易纯化及环境友好等优点.     

陕西省长寿区老人血清中锰、铜、锌、铁及钙、镁的分析

作者对陕西省长寿区老人血清中微量元素(锰、铜、锌、铁)及钙、镁进行分析。发现陕西省长寿区老人血清中利于健康长寿的锰的含量极显著地高于对照区(P<0.001);人体必需的铜的含量也极显著地高于对照区(P<0.01);与抗衰老关系密切的钙的含量也极显著地高于对照区(P<0.01);男性老人血清中锰、铜、锌、铁及钙、镁的含量与女性老人均无显著差异;61岁～90岁的老人血清中锰、铜、锌、铁及钙、镁的含量随着年龄的增长也均未有显著差异。结果表明陕西省长寿区老人血清中有较优越的化学元素谱。本文对实验结果中与抗衰老有关问题进行了讨论。     

磷酸铵镁技术中不同沉淀剂脱氮除磷效果比较

探讨了磷酸铵镁法中不同沉淀剂的脱氮除磷效果,比较了氨氮去除率、磷酸盐去除率、残氮量、残磷量,沉淀物质质量和组成,处理液的电导率。综合考虑了各方面因素得出处理高浓度氨氮废水的较佳的沉淀剂为Na3PO.412H2O MgCl2.6H2O和Na3PO4.12H2O MgSO4.7H2O,处理高浓度磷酸盐废水的较佳的沉淀剂为NH4Cl MgCl2.6H2O和NH4Cl MgSO.47H2O.并对磷酸铵镁沉淀进行了X-衍射光谱和扫描电镜表征。

镁,在当前的加工制造业中正扮演着越来越重要的角色,是世界所公认的极重要的有色金属之一。镁合金具有比重轻、比强度和比刚度高、导热导电性好、兼有良好的阻尼减震和电磁屏蔽性能、易于加工成型、容易回收等优点。它比铝高30倍的减振性能、比塑料高200倍的导热性能、其热膨胀性能     

矿物微粉和憎水剂对磷酸镁水泥基材料耐水性影响的试验研究

为了改善磷酸镁水泥基材料的耐水性能,通过掺加矿物微粉和憎水剂对磷酸镁水泥基材料的强度保留率、电通量和膨胀率进行研究。结果表明:掺加了矿物微粉和憎水剂的磷酸镁水泥基材料强度保留率较基准有所增加,矿物微粉掺量为20%时,各龄期强度保留率达到了最大值;在标准养护和水养的条件下,随着矿物微粉掺量的不断增加,磷酸镁水泥基材料的电通量呈逐渐降低趋势;矿物微粉掺量为20%时,28d电通量达到850C和900C;掺加20%矿物微粉和0.3%憎水剂的磷酸镁水泥基材料的膨胀率要高于基准组0.05%。
