DNA序列分析确认1例新的HLA-B等位基因B4814

目的识别确认中国人群中1例新的HLA等位基因。方法使用PCR-SSP和测序为基础的分型技术,分析确认HLA-B*的新等位基因。结果先证者HLA-B位点有一个等位基因的核苷酸序列与已知的HLA等位基因均不同,该基因和B*4812的差异在第3外显子区域的419位碱基A>C,486位碱基C>G,512位碱基G>T。导致116编码子TAC>TCC,编码的氨基酸由酪氨酸(Tyr)变成丝氨酸(Ser),147编码子TGG>TTG,编码的氨基酸由色氨酸(Trp)变成亮氨酸(Leu)。结论该基因为HLA新的等位基因,2005年10月被世界卫生组织HLA因子命名委员会正式命名为HLA-B*4814。     

平阳霉素局部注射对鼻息肉病息肉组织病理学的影响

目的通过观察平阳霉素治疗前后鼻息肉病息肉组织的病理变化情况,探讨平阳霉素治疗鼻息肉病的机制。方法应用末端脱氧核苷酸转移酶介导的脱氧三磷酸尿苷缺口末端标记法(TUNEL法)检测凋亡细胞,与相邻连续的HE染色片对位对照观察嗜酸性粒细胞的凋亡情况。结果平阳霉素治疗前鼻息肉组织嗜酸性粒细胞表达显著高于正常下鼻甲(P<0.01);治疗后鼻息肉组织中的嗜酸性粒细胞表达明显低于治疗前(P<0.01);治疗后鼻息肉组织中嗜酸性粒细胞的凋亡指数高于治疗前(P<0.01)。结论嗜酸性粒细胞是鼻息肉病息肉组织中主要的浸润炎细胞;平阳霉素可通过促进嗜酸性粒细胞凋亡达到治疗鼻息肉病的目的。     

β淀粉样肽1～42单克隆抗体的制备

目的　制备稳定分泌β -淀粉样肽 1～ 4 2 (Aβ1-42 )单克隆抗体的杂交瘤细胞 ,产生高效价抗体。方法　通过基因工程技术 ,将Aβ1-42 基因融合于乙肝核心抗原 (HBcAg)的主要免疫优势区 (MIR) ,制备Aβ和HBcAg的融合蛋白 ,免疫BALB/c小鼠 ,取脾细胞与SP2 / 0细胞融合 ,筛选能稳定分泌Aβ1-42 单克隆抗体的杂交瘤细胞。结果　得到两株能稳定分泌Aβ1-42 单克隆抗体的杂交瘤细胞 ,Aβ1-42 单克隆抗体的Ig亚类为IgG3 。结论　制备得到的Aβ1-42 单克隆抗体特异性强 ,效价高     

基于双层规划的公交车调度问题的模型与算法

城市公交车辆调度的主要目的是在方便乘客出行的前提下追求企业利益的最大化,其关键问题是在企业如何进行公交车辆配置和安排一天中各个时段上的发车频率.在对现有问题研究的基础上,基于对调查数据的分析和推导,建立了以乘客满意度为上层目标、企业运营效益为下层目标的双层规划模型,并对此设计了一个遗传模拟退火混合优化算法,结果表明:本文提出的双层模型和设计的遗传模拟退火算法具有一定得有效性和可行性.
