β淀粉样肽1～42单克隆抗体的制备

目的　制备稳定分泌β -淀粉样肽 1～ 4 2 (Aβ1-42 )单克隆抗体的杂交瘤细胞 ,产生高效价抗体。方法　通过基因工程技术 ,将Aβ1-42 基因融合于乙肝核心抗原 (HBcAg)的主要免疫优势区 (MIR) ,制备Aβ和HBcAg的融合蛋白 ,免疫BALB/c小鼠 ,取脾细胞与SP2 / 0细胞融合 ,筛选能稳定分泌Aβ1-42 单克隆抗体的杂交瘤细胞。结果　得到两株能稳定分泌Aβ1-42 单克隆抗体的杂交瘤细胞 ,Aβ1-42 单克隆抗体的Ig亚类为IgG3 。结论　制备得到的Aβ1-42 单克隆抗体特异性强 ,效价高     

瘦素受体在大鼠中枢神经系统的免疫组织化学定位研究

目的　观察瘦素受体在SD大鼠中枢神经系统的组织学定位。方法　采用高度特异的抗瘦素受体血清 ,用免疫组织化学ABC法观察瘦素受体在大鼠脑的分布。结果　在SD大鼠的前脑 (包括嗅球 )、皮层Ⅳ、Ⅴ层、海马、下丘脑、杏仁核、丘脑等可以观察到瘦素受体免疫反应阳性物质 (LR IR) ,其中下丘脑弓状核、视上核、视交叉上核、杏仁基底外侧核、孤束核有较强的LR IR ,而下丘脑外侧视前区、杏仁中央核有较弱的LR IR。结论　瘦素受体广泛存在于大鼠中枢神经系统 ,包括下丘脑、杏仁核等与味觉和摄食调节密切相关的核团     

环氧化酶-2及表皮生长因子受体在非小细胞肺癌组织中的表达及临床意义

目的探讨环氧化酶-2(COX-2)与表皮生长因子受体(EGFR)在非小细胞肺癌(NSCLC)组织中的表达及与肺癌发生发展的关系。方法采用免疫组化(SP)法检测56例NSCLC和20例正常肺组织中COX-2与EGFR的表达。结果56例NSCLC组织中,COX-2与EGFR表达阳性率分别为64.2%和67.8%,显著高于正常肺组织(P<0.01),COX-2与EGFR表达与临床分期、病理分级、组织学分型和淋巴结转移密切相关(P<0.05),肿瘤分期愈晚,细胞分化越差,淋巴结转移越高,COX-2与EGFR表达阳性率越高,腺癌中COX-2表达明显高于鳞癌,鳞癌中EGFR表达高于腺癌(P<0.05)。结论COX-2与EGFR在NSCLC中异常表达说明其在肺癌发生发展中有重要作用,可作为判断NSCLC的生物学行为及估计预后的指标。

肺癌患者外周血T细胞亚群及免疫细胞活性的研究

采用 APAAP法和流式细胞仪研究 32例肺癌患者及 30例健康人外周血T细胞亚群及免疫细胞活性 ( NK- A)的变化。结果表明 :肺癌患者外周血 CD+3 、CD+4、CD+4/CD+8及 NK- A与正常人比较呈显著性降低 ( P <0 .0 5 ,P <0 .0 1 ) ;且肺癌伴癌转移者与未转移者上述各项指标亦有显著性差异 ( P <0 .0 5 )。说明肺癌患者存在细胞免疫功能降低 ;伴有癌转移的肺癌患者细胞免疫功能更低。提出积极调节肺癌患者细胞免疫功能 ,增强机体抗肿瘤能力十分重要。
