成人界嵴形态与组织结构的观察   总被引:3,自引:0,他引:3  
目的 观察和测量人右心房界嵴和梳状肌的形态和组织结构 ,以探讨其可能的功能。方法 肉眼观察、测量界嵴与梳状肌的有关形态学数据。利用光学显微镜 ,通过HE、Masson和磷钨酸 苏木素染色 ,对界嵴的细胞形态和纤维方向等进行细致观察。结果 ①根据界嵴的隆起状况 ,界嵴可分为 4类 :A .全程隆起 (18.6 % ) ;B .上 2 / 3隆起 ,下1/ 3平坦 (2 7.9% ) ;C .上 1/ 3隆起 ,下 2 / 3平坦 (34.88% ) ;D .全程平坦 (18.6 % )。②界嵴的肌纤维可分深、浅两层 ,深层肌纤维纵行 ,与界嵴长轴平行 ;浅层纤维横行 ,仅见于界嵴的中、下部 ,深 浅层纤维之间互相联系。在界嵴各处可见P样细胞。③根据梳状肌的起止和走行 ,梳状肌可分为 3型 :汇聚型、网状型和混合型。结论 窦房结上、下及其深面借过渡纤维与界嵴的普通肌纤维相互联系 ,界嵴纤维可能与结间冲动传导有关。梳状肌走行方向的多样性 ,可能与临床上心脏电流传导的多向异性有关  相似文献

Objective Ependymal cells are thought to be the primary source of neural stem cells in the adult central nervous system. The purpose of this study is to examine spatial and temporal profiles of ependymal cell proliferation and migration after focal cerebral ischemia. Methods Eighty male Sprague Dawley rats underwent permanent middle cerebral artery occlusion after injection of 10/μL of 0.2% Dil into the lateral ventricle. Rats were sacrificed and brain sections were acquired for pathological evaluation and laser confocal imaging at day 1,3,7,11,14,21 and 28 after ischemia. Results The density of Dil-labeled cells in the ischemic ipsilateral subventricular zone was significantly higher than that in the control group and these labeled cells dispersed in the ischemic ipsilateral subventricular zone and/or were located in ependyma from day 1 to 11. In the ischemic ipsilateral cortex, some Diilabeled cells occurred in peri-infarction and infarction of parietal region at day14 and peaked at day 21 when some Dil-labeled cell nodules were found in this region. During postischemic day 14--28, a significant decrease in labeled celldensity in the ischemic ipsilateral subventricular zone was coincident with a significant increase in labeled cells density in the cortex (peri-infarction and infarction). Conclusion The results indicate that ependymal cells proliferate and migrate after focal cerebral ischemia in the adult rat brain.  相似文献

中药内异消对大鼠子宫内膜异位症血管生成因子的影响   总被引:11,自引:1,他引:11  
目的 探讨血管内皮生长因子 (VEGF)、肿瘤坏死因子 (TNF α )在子宫内膜异位症 (EM)发病机制中的作用及相互关系 ,观察中药内异消对两因子的影响及对EM的治疗作用。方法 采用种植内膜方法建立EM动物模型 ,内异消治疗 ,丹哪唑对照 ,ELISA法测定TNF α水平 ,免疫组化SP法检测VEGF表达。结果 EM病模组异位内膜组织中VEGF的表达及腹腔液TNF α含量显著增高 ,与假模型组比较有统计学差异 (P <0 .0 1) ;内异消能降低VEGF表达及TNF α含量 ,与病模组比较有统计学差异 (P <0 .0 1)。结论 VEGF、TNF α在EM发生发展中起重要作用 ,内异消可通过调节两因子的表达来抑制异位内膜的生长  相似文献

Objective To investigate the mechanisms of apoptosis induced in Human leukemia cell line K562 by the combination of indole-3-acetic acid and horseradish peroxidase. Methods Human leukemia cell line K562 were exposed to indole-3-acetic acid (IAA) at 20, 40, 60, 80 or 100 mol/L and horseradish peroxidase(HRP) at 1.2 g/mL for varying times. MTT assay was applied to detect the cell proliferation. Flow cytometry was performed to detect the arrest of cell cycle. Terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay was used to measure apoptosis. 2, 7-dichlorofluorescin diacetate (DCFH-DA) uptake was measured to determine free radical by confocal microscope. Content of malondiadehyde (MDA) and activity of superoxide dismutase (SOD) were measured by biochemical methods. Results IAA/HRP initiated growth inhibition of K562 cells in a dose- and time-dependent manner. Flow cytometry revealed that cell cycle arrested at G1/G0 after 24 hours treatment. After 72 hours treatment, apoptotic rate of 100 mol/L IAA group increased to 43.9 %, which was 5 times that of control(P〈0.01). Content of MDA and activity of SOD increased respectively in treatments compared with control. Meanwhile, IAA/HRP stimulated the formation of free radical, which was increased by IAA concentration-dependently. Conclusion The combination of IAAand HRP can inhibit the growth of Human leukemia cell line K562 in vitro by inducing apoptosis which is associated with the increase of free radical. The combination of IAA and HRP might be a promising chemopreventive and chemotherapeutic agent against human leukemia.  相似文献

目的 研究尾部悬吊对大鼠比目鱼肌梭内、外肌纤维的mATP酶活性的影响 ,旨在探讨失重条件下梭内、外肌代谢的改变。方法 采用尾部悬吊法建立模拟失重模型 ,以钙增强的mATPase法检测模拟失重不同时期大鼠比目鱼肌梭内、外肌的mATP酶活性。结果 尾部悬吊可使梭外肌Ⅰ型肌纤维的构成比减少 ,Ⅱ型肌纤维的构成比增加 ,梭内肌纤维mATP酶活性增强 ,核袋 1纤维mATP酶染色不变或由阴性转变为弱阳性 ,核袋 2纤维和核链纤维由阳性转变为强阳性。结论 模拟失重导致梭内、外肌代谢发生改变 ,梭外肌纤维类型发生转化。  相似文献

Objective To investigate the properties of proliferation and differentiation of neonatal rat retinal progenitor cells (RPCs) in vitro. Methods RPCs were isolated from neonatal SD rats neural retina and cultured in DMEM/F12 + N2 with EGF and bFGF (suspension medium ) or 10 % FBS without EGF and bFGF ( differentiation medium). The cells grew as suspended spheres or adherent monolayers, depending on different culture conditions. The neural stem cells or retinal progenitors, neurons, astrocytes, retinal ganglion cells, rod photoreceptors and the proliferating cells were evaluated with immunofluorescence analysis by Nestin or Pax6, Map2, GFAP, Thy-1, Rhodopsin and BrdU antibodies respectively. Results RPCs could propagate and differentiate in suspension or differentiation medium and express the markers of Nestin (92.86%) or Pax6 (86.75%), Map2 (38.54%), GFAP (20.93%), Thy-1 (27. 66%) and Rhodopsin(13. 33%)in suspension medium; however, Nestin (60.27%), Pax6 (52%), Map2 (34.94%), GFAP (38.17%), Thy-1 (30.84%) and Rhodopsin (34.67%) in differentiation medium. 96.4 % of the population in the neurospheres was BrdU-positive cells. The cells could spontaneously adherent forming some subspheres and retinal specific cell types. Conclusion Neonatal rat RPCs possess the high degree of proliferation and can differentiate into neurons, astrocytes, retinal ganglion cells and rod photoreceptors in vitro. There are different proportions for RPCs to differentiate into specific cell types.  相似文献

目的探讨苦参素(Matrine)对体外培养的人宫颈癌Hela细胞增殖及凋亡的影响。方法Hela细胞体外培养,MTT法检测细胞增殖,TUNEL法检测细胞凋亡,免疫组化法检测细胞增殖核抗原(PCNA)蛋白的表达水平。结果与对照组相比,0.5、1.0、1.5、2.0、2.5、3.0g/L苦参素均能显著抑制HeLa细胞生长(P<0.05),药物浓度在0.5-1.5g/L之间时表现出时效和量效关系;苦参素2.0、2.5、3.0g/L与1.5g/L相比,抑制无统计学意义(P>0.05)。与对照组相比,1.0、1.5、2.0g/L苦参素组可见到显著性细胞凋亡(P<0.05);1.5g/L苦参素组作用48h和72h时PCNA蛋白表达显著下调(P<0.05)。结论苦参素可能通过抑制增殖和诱导凋亡的双重途径抑制宫颈癌Hela细胞的生长,苦参素有可能成为治疗子宫颈癌的一种药物。  相似文献

目的了解转化生长因子-β2(TGF-β2)对体外培养的牛眼小梁(TM)细胞基质金属蛋白酶-2(MMP2)的表达和活性的影响,探讨TGF-β2在原发性开角型青光眼(POAG)发病机制中的作用。方法用含有1.0μg/L TGF-β2或含1.0μg/L TGF-β2+100g/L鼠抗人PAI-1IgG(PAI-1中和抗体)的无血清DMEM分别孵育TM细胞24、36、48h。对照组(Co)加入不含实验药物的无血清DMEM,用Western印迹法检测MMP2及纤溶酶原活化抑制剂-1(PAI-1)的表达。结果TGF-β2可显著增强TM细胞MMP2前体及PAI-1的表达。PAI-1中和抗体可促进MMP2前体向其活化形式转化。结论TGF-β2可通过抑制MMP2的活化过程导致TM细胞的细胞外基质(ECM)的异常堆积,造成房水引流阻力的增加,参与POAG的发病。  相似文献
