Rapid quantification of the metabolite of valacyclovir hydrochloride in human plasma by liquid chromatography-tandem mass spectrometry

Objective To establish a rapid, sensitive and selective liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the determination of acyclovir (the metabolite of valacyclovir hydrochloride) in human plasma. Methods After addition of ganciclovir as internal standard (IS), plasma samples were prepared by one-step protein precipitation using acetonitrile as precipitant, followed by an isocratic elution with 0.1% formic acid 3.5μm) column. Detection was performed on a triple-quadrupole mass spectrometer utilizing electrospray ionization (ESI) interface operating in positive ion and selected reaction monitoring (SRM) mode with the precursor to product ion transitions m/z 226.2→152.1 for acyclovir and m/z 256.2→152.1 for the IS. Results The analytical results demonstrated a good linearity over the ranges from 0.005 to 4μg/mL (r=0.9999) for valacyclovir hydrochloride. The relative standard deviations (RSD) of intra-batch and inter-batch were less than 4.06% and 9.23%, respectively. The limit of detection and lower limit of quantification in human plasma were 2ng/mL and 5ng/mL, respectively. Conclusion The method was simple, sensitive, accurate and reproducible and has been successfully applied to a bioequivalence study of valacyclovir hydrochloride capsules in Chinese healthy male volunteers.     

基于RBF神经网络的TCSC自适应PID控制

针对可控串联补偿(Thyristor Controlled Series Capacitor,TCSC)的传统PID控制器存在鲁棒性差和自学习能力不强的问题,利用径向基函数(Radial Basis Function,RBF)神经网络可以无限逼近非线性系统的特性,提出了一种基于RBF神经网络设计的TCSC自适应PID控制器.该自适应PID控制器不仅具有传统PID控制器结构简单、易于工程实现等特点,而且通过RBF神经网络辨识的被控系统Jacobian信息来在线实时调整PID控制的参数,对被控系统参数和运行状态改变有较强的适应性,克服了常规PID控制器鲁棒性不强的缺点.两机五节点系统和四机两区域系统的仿真结果表明所设计的控制器控制效果明显优于传统PID控制器,具有较好的适应性和鲁棒性,对系统低频振荡的阻尼特性有很大地改善.     

3D numerical modeling of wave forces on tandem fixed cylinders using the BEM

In this paper a 3D numerical model was developed to study the complicated interaction between waves and a set of tandem fixed cylinders.The fluid was considered to be inviscid and irrotational.Therefore,the Helmholtz equation was used as a governing equation.The boundary element method（BEM） was adopted to discretize the relevant equations.Open boundaries were used in far fields of the study domain.Linear waves were generated and propagated towards tandem fixed cylinders to estimate the forces applied on them.Special attention was paid to consideration of the effect on varying non-dimensional cylinder radius and distance between cylinders,ka and kd on forces and trapped modes.The middle cylinder wave forces and trapped modes in a set of nine tandem cylinders were validated utilizing analytical data.The comparisons confirm the accuracy of the model.The results of the inline wave force estimation on n tandem cylinders show that the critical cylinder in the row is the middle one for odd numbers of cylinders.Furthermore the results show that the critical trapped mode effect occurs for normalized cylinder radiuses close to 0.5 and 1.0.Finally the force estimation for n tandem cylinders confirms that force amplitude of the middle cylinder versus normalized separation distance fluctuates about that of a single cylinder.

V形沟谷加筋土支挡结构强度特性分析

笔者研究的加筋土支挡结构所用筋带为钢筋砼串联块，此结构修筑V形冲沟中，作者采用C、φ值同时提高的方法分析了此结构的强度特性，推导了加筋土在两种破坏模式下的墙顶部表面承载力，从而表明此种结构物具有广阔的应用前景。
