动车运用所室外综合管线是指所内室外各个专业的管沟、管槽、管线、管廊等,其数量种类多、交叉点数量庞大、多专业设计内容又互为边界条件,综合管线设计一直是动车所工程的重难点。传统二维施工图设计具有局限性,协调难度大,容易造成施工阶段管线安装工序不当,进而引发安装空间不足或管线冲突等问题。提出基于BIM的三维正向设计方法,通过研究协同设计、重要基准导入、三维管线综合排布方法、复杂交叉点优化方法、管线碰撞检查和调整方法等,立体直观地展现综合管线的空间排布位置,同时优化三维模型转二维施工图出图方法,合理规划出图内容,确保图纸表达完整,减少设计出错率,指导施工,减少返工和浪费人力物力等现象,为后续复杂段所室外综合管线正向设计提供借鉴。  相似文献

Nowadays, bulbous bow forms have become a common design feature in most conventional ship designs. The design of a bulbous bow is usually attempted with the use of certain form parameters that are imposed using the designer's experience or regression values that provide the optimum parameters based on experimental tests or computational fluid dynamics (CFD) calculations. In this article, the geometric modelling of a bulbous bow form that complies with a series of parameters is presented. First, a wire model was constructed with cubic B-spline curves that hold certain form parameters that the designer wants to impose. Second, a B-spline surface that fits these splines was mathematically constructed. A review of the influence of the design parameters used on the bulbous bow properties was made. This method could be used prior to numerical optimization of a bulbous bow because different variations of the design can be easily generated. These variations can be tested numerically using CFD software. A practical application example of the method is shown.  相似文献

合理布置桥区航道对于船舶在桥区的安全航行非常重要.桥墩的紊流范围是布置桥区航道时要考虑的一个重要指标.文中分析了顺直航道圆形桥墩紊流范围的相关主控因素.通过设置主控因素的序列数据,数值模拟计算并经回归分析,得出了顺直航道圆形桥墩最大紊流范围的经验表达式.研究结果表明,桥墩最大紊流范围与水面流速和桥墩直径密切相关,且流速和桥墩直径越大,桥墩紊流最大范围也越大.  相似文献

目的对作者发现的一个中国3代人心脏传导阻滞家系SCN5A基因新突变L1001Q,构建突变表达载体,利用分子生物学方法和全细胞膜片钳技术观察L1001Q突变的功能。方法一步定点诱变法构建pEGFP-L1001Q-SCN5A突变体;激光共聚焦和Western blotting技术检测突变蛋白定位和表达;全细胞膜片钳技术观察突变体钠电流。结果野生型和L1001Q突变均表达在细胞膜上,且表达量无明显变化。二者最大激活电流分别为(148.34±0.77)pA/pF和(132.59±0.96)pA/pF(P>0.05),L1001Q突变半失活电压V1/2为(-73.25±0.87)mV,较野生型正向转移约3.5mV[V1/2为(-76.71±0.73)mV,P<0.05],L1001Q突变失活后恢复时间常数较野生型稍减小,差异无统计学意义(WT tau=16.8ms,L1001Qtau=14.1ms,P>0.05)。结论 L1001Q突变未影响钠通道蛋白的表达和转运,主要通过改变钠通道门控特性引起钠通道功能减弱进而导致心脏传导阻滞。  相似文献

Objective To investigate the expression of neuroprotective peptide [Gly14]-Humanin (HNG) in eukaryotic cells by gene engineering technique and analyze its biological activity. Methods By means of asymmetrical primer/template, double stranded cDNA of HNG with FLAG in its C-terminal was obtained, which was cloned into the plasmid pcDNA3.1(-), and the resultant recombinant vector pcDNA3.1(-)/HNG-FLAG was transfected into PC12 cells. At the same time, the recombinant vector pcDNA3.1(-)/EGFP was transfected to control the efficiency of transfection. The expression of HNG in the cells was determined by immunocytochemistry. In order to analyze the biological activity of the expressed HNG, 25μM Aβ25-35 peptide was added to the culture medium of the transfected cells for 24h, then cell morphology, MTT assay and Hoechst 33258 staining were observed. Results The eukaryotic expression vector of pcDNA3.1(-)/HNG-FLAG was identified by enzyme digestion and sequencing. HNG was highly expressed in PC12 cells. After exposure of PC12 cells to 25μM Aβ25-35 for 24h, cell viability decreased to (65.8±5.3)%, and the dystrophic changes of neuritis and nuclei condensation were obvious. When cells were pre-transfected with pcDNA3.1(-)/HNG-FLAG, Aβ25-35-induced cell death and morphological changes of cells and nuclei were suppressed. In contrast, pre-transfected with empty vector did not protect cells from Aβ25-35-induced toxicity. Conclusion The eukaryotic expression vector for FLAG-tagged HNG was successfully constructed and expressed in PC12 cells. Expressed HNG has biological activity.  相似文献

In this paper, a focusing approach is presented to widen the use of efficient monostatic imaging algorithms for azimuth-invariant bistatic synthetic aperture radar (SAR) data. The bistatic range history is modeled by a polynomial of azimuth time. Using this model, an analytic form of the signal spectrum in the 2D frequency domain is derived, and a simple single-valued relation between the transmitter and receive ranges is established. In this way, a lot of monostatic image formation algorithms can be extended for the bistatic SAR data, and a bistatic chirp scaling algorithm is developed as an application of the new approach. This algorithm can be used to process the azimuth-invariant bistatic configuration where the transmitter and receiver platforms are moving on parallel tracks with the same velocity. In addition, some simulation results are given to demonstrate the validity of the proposed approach.  相似文献

基因表达式编程作为新型的人工智能数学建模方法,具有较强的函数发现能力和较高的搜索效率.针对公路工程造价预测特点和已有预测模型的不足,求解基于GEP的公路工程造价预测模型.根据工程造价的组成原理筛选确定11个公路工程特征因素,用15组公路工程数据作训练样本和验证样本,在设定好GEP算法的构成要素的前提下,使用Visual Basic语言表达GEP算法,求解公路工程造价预测模型.通过敏感性分析模型的可行性,用比较图和4个统计指标值对模型进行评价.结果表明,预测模型所得结果与工程实践相吻合,且预测值与实际值的相对误差≤5.9%,满足预测精度≤10%的要求,预测精度高,该模型具有较好的应用价值.  相似文献

目的 构建带有 6×myc的人白介素 17受体样分子的重组表达质粒 ,以便检测hIL 17RLM L基因在真核细胞中的特异性表达。方法 设计带有酶切位点 (EcoRⅠ和XhoⅠ )的特异性引物 ,用PCR方法扩增hIL 17RLM L片段回收后插入带有 6×myc标签的 pcDNA3.0真核表达载体 ,转染COS7细胞后作Westernblot检测其表达。结果 成功地构建了带有 6×myc标签的 pcDNA3.0 6×myc /hIL 17RLM L重组质粒 ,Westernblot检测到该质粒可在真核细胞中表达。结论 用分子生物学的方法成功地构建了真核表达质粒pcDNA3.0 6×myc /hIL 17RLM L ,使该基因的特异性检测成为可能 ,为进一步研究hIL 17RLM L的生物学功能奠定了基础。  相似文献
