肿瘤细胞E-cadherin的表达与对基底膜浸润能力的关系

摘    要:目的 了解E cadherin下调对肿瘤细胞侵袭能力的影响。方法 选用人胰腺癌细胞株JHP 1、PANC 1、MIAPaCa 2 ,运用免疫细胞化学染色、Westernblot及细胞浸润MTT比色法 ,了解细胞E cadherin蛋白表达情况及该细胞对基底膜的浸润。结果 免疫细胞化学染色提示 ,E cadherin表达位于细胞膜。JHP 1细胞株表达较强 ,PANC 1着色减弱 ,MIAPaCa 2几乎无着色。Westernblot表明E cadherin蛋白JHP 1含量最高 ,PANC 1次之 ,而MIAPaCa 2含量不易测出。细胞浸润MTT比色法确定MIAPaCa 2浸润能力最强 ,PNAC 1次之 ,JHP 1最弱 ,经统计学处理差异显著 (P <0 .0 1)。结论 E cadherin不仅具有介导细胞间粘附的作用 ,而且与肿瘤细胞的浸润能力有关。

Abstract:Objective To study the effect of down-regulation of E-cadherin on the invasive ability of tumor cells. Methods Human pancreatic carcinoma cell lines JHP-1, PANC-1, and MIA PaCa-2 were selected. The immunocytochemistry, Western blot and invasive-MTT assay were used to observe the expression and the contents of E-cadherin in the tumor cells. Results Immunocytochemistry showed that E-cadherin expression was on cell membrane. The expressions of E-cadherin were preserved in JHP-1, reduced in PANC-1, and showed negative in MIA PaCa-2. Western blot analysis showed that the protein content was the highest in JHP-1 and lowest in MIA PaCa-2. According to invasive MTT assay, the invasive ability of MIA PaCa-2 was the strongest, followed by PANC-1 and JHP-1 (P< 0.001). Conclusion E-cadherin is not only an adhesive molecule that mediates adhesion of cells, but also related to the invasive ability of tumor cells.
