高速放射自显影神经追踪术探讨

摘    要:用氚标记的氨基酸进行放射自显影研究需要的曝光时间太长。一般当切片蘸乳胶以后需要在冰箱内置暗盒中曝光2~7周。国内有些研究者为了提前检查氚标记物的注射区曾使用过用闪烁液加速曝光的办法,但从来没有见到用闪烁液处理全部切片进行神经追踪的研究。

Abstract:3、 5、 10、20 μci of ~3H-Lucine were injected into the olfactory bulb of 13 rats, the survival time was 2、4、6 days respec- tively.Having been fixed by 10% forma- lin,the brain of the animals was embeded in paraffin and cut serially.(3 cases by cryostat section).The sections scintillated in the scintillate solution then were expo- sured 4 hours after dipped in emulsion. The results arc as follows:1.The high speed autoradiography might be used to trace the nervous patbway;2.Scintillating once,together with exposuring 4 hours is sufficient to expose siliver grain in the most distant region;3.10 μci tracer is a desirable dosage if injecting tracer in nucl- eus of brain;4.4~6 days is the best sur- vival time;5.The result of cryostat sec- tions of 3 cases can also be used to study ARG.The high speed autoradiographic nerve tracing can shorten the period of reseach from 5~7 weeks to 5~7 days.
