硒、维生素E对自由基损伤培养兔软骨细胞的保护作用

摘    要:采用细胞培养方法,以黄嘌岭-黄嘌岭氧化酶(XXO)系统产生自由基,造成软骨细胞的自由基损伤;软骨细胞DNA及蛋白多糖的合成受到明显抑制;膜流动性降低;电镜下观察到细胞表面有大小形态各异的囊泡状突起,胞内可见线粒体肿胀或呈固缩样改变、溶酶体增多等超微病理损害。向培养基中提前1d加入硒及维生素E能对抗自由基所致的氧化性损伤。

Abstract:In this paper, using the method of the rabbit articular chondrocytes (RACs) in the monolayer culture in vitro, we studied the effect of superoxide free-radical generated by the xanthine-xanthine oxidase (XXO) system on RACs. The results showed that oxygen free-radicals inhibited the synthesis of RACs' DNA and proteoglycan, decreased membrane fluidity. The chondrocytes grew slowly and were not confluent under microscope af-ter six days. The changes of cellular ultrastructural damage under TEM and SEM were observed clearly. Selenium or vitamin E added into the medium 24 hours before adding the XXO system was able to protect RACs from being injured by free radicals. These results suggest that selenium or vitamin E may play an important role in retarding the process of cartilage disease.
