摘    要:本文介绍3.3'、5.5'—四甲基联苯胺(TMB)作为辣根过氧化物酶(HRP)的底物应用于ELISA检测乙型肝炎表面抗原(HB_sAg)。TMB敏感性高、反应产色鲜明、本底反应低、终止后稳定,用于检测HB_sAg的ELISA系统,使敏感性比目前最常用的底物邻苯二胺(OPD)提高了8~16倍。TMB是一个比较理想的HRP底物,用于检测HB_sAg的ELISA系统可以提高阳性检出率,有一定的实用价位。

Abstract:3,3,'5,5'-tetramethyl-benzidine(TMB) a new substrate of horse-radish peroxidase(HRP) has been used in ELISA for the detection of Hepatitis B surface antigen(HBsAg)in sera from patients with Hepatitis B and health persons. TMB's main advantages arc noncarcinogenie,highly sensitive, and rather stable. The optimum reactive conditions of TMB in this experiment has been determined. TMB as the substrate of HRP could increase the sensitivity of ELISA for detection HB~sAg for 8 to 16 times more than the ordinary subatrate of HRP, O-Phenylenediamine(OPD), and the positive detecting rate of HB_sAg for about17per cent.TMB is a better substrate of HRP at present.
