摘    要:目的 探讨表皮生长因子 (EGF)在脑膜瘤细胞中的表达和作用 ,以及体外针对该生长调节环路抑制脑膜瘤细胞增殖的可能性。方法 通过对 2 1例脑膜瘤原代培养 ,采用ABC免疫细胞化学法检测EGF和EGF受体 (EGFR)在脑膜瘤细胞中的表达 ,并选用四唑盐 (MTT)比色法观察EGF、EGF单抗和EGFR单抗对脑膜瘤细胞的增殖调控作用。结果 脑膜瘤细胞可表达EGF和EGFR ;EGF以剂量依赖关系刺激培养脑膜瘤细胞增殖 ,而EGF单抗和EGFR单抗皆以剂量依赖关系抑制培养脑膜瘤细胞 ,且EGFR单抗的抑制效果明显强于EGF单抗 (P <0 .0 1)。结论 EGFR单抗较EGF单抗是更为有效的脑膜瘤增殖抑制单抗 ,为脑膜瘤的生物治疗提供新的实验基础

Abstract:Objective To investigate the expression and function of epidermal growth factor (EGF) in human meningioma cells, and the possibility of inhibiting the proliferation of human meningioma cells in vitro by interrupting the proliferation-stimulated cycle. Methods Meningioma tissues from 21 cases were cultured. ABC immunocytochemical method was used to examine the expression of EGF and epidermal growth factor receptor (EGFR) in human meningioma. MTT assay was applied to evaluate the effect of EGF,EGF monoclonal antibody and EGFR monoclonal antibody on the proliferation of cultured meningioma cells. Results EGF and EGFR were expressed by human meningioma; EGF stimulated, while EGF monoclonal antibody and EGFR monoclonal antibody inhibited the proliferation of cultured human meningioma cells, in a dose-dependent manner, and the maximal inhibitory effect induced by EGFR monoclonal antibody was much stronger than the effect caused by EGF monoclonal antibody. Conclusion Both EGF monoclonal antibody and EGFR monoclonal antibody inhibit the proliferation of cultured meningioma cells, providing a new therapeutic basis for biological therapy of meningiomas.
