| 摘 要： | 本实验比较了新鲜分离的正常人外周血单个核细胞(PBMC)和佛波脂类促癌剂TPA活化的PBMC对重组人白细胞介素4(γIL_4)促增殖作用的应答。发现γIL_4可剂量依赖性地促进新鲜分离的PBMC增殖,但其程度太小不足以灵敏反应γIL_4量的变化,TPA可激活PBMC,从而提高PBMC中活化细胞比例,活化的PBMC对γIL_4刺激的应答增强,细胞增殖与rIL_4浓度之间存在较明显的正相关性。因此TPA活化的PBMC的增殖反应可作为一种较为简便,灵敏的γIL_4定量检测方法。

| Abstract: | In this paper, the respones of freshly seperatedperipheral blood mononuclear cells(PBMC) and TRAactivated PBMC to the promoting--proliferation effectsof recombinant human interleukin--4(γIL--4) werecompared. We discovered γIL--4 could promoted fresh-ly seperated PBMC from healthy qonors proliferation ina does--dependent fashion. but the proliferation extentwas too small to show the change of γIL--4 concentra-tion. TPA could activate PBMC and raised the ratio ofactivated cells. The activsaed PBMC showed enhancedproliferation response to γ7IL--4 stimulation and therewas a clear correlation between cells proliferation andγIL--4 concentration. So the proliferation response ofTPA activated PBMC could be considered as an casyand a sensitive method for γIL--4 assay. |
