摘    要:目的应用液体芯片-飞行时间质谱技术从慢性阻塞性肺疾病(COPD)患者的血清中找出差异表达蛋白,筛选出其蛋白多肽(m/z)的疾病标志物。方法应用液体蛋白芯片(MB-WCX)结合飞行时间质谱(MALDI-TOF-MS)技术对16例病理确诊的COPD患者和32例正常对照人群进行血清蛋白组分析。采用ClinProTools软件进行蛋白多肽的组间差异对比,共获得115个蛋白多肽峰图,其中24个蛋白多肽峰图具有极显著差异(P<0.001),其中10个蛋白多肽在COPD患者中表达下调,其余14个蛋白多肽在COPD患者中表达上调。两组数据中最大差异的两个蛋白多肽峰分别是m/z:1 012.92和m/z:4 152.85。结论利用液体芯片-飞行时间质谱技术可以从COPD患者的血清中获得稳定丰富的蛋白多肽图谱,并且能够筛选出潜在的疾病标志物。

Abstract:Objective To screen potential specific biomarkers in serum of chronic obstructive pulmonary disease(COPD) patients using pre-fractionated magnetic bead based sample fractionation method combined with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS) technology.Methods We compared serum proteomic profiling of 16 COPD patients and 32 healthy controls by MB-WCX-MALDI-TOF-MS.Results By analyzing the spectra(screened from two groups) using the ClinProTools software,24 of all 115 peaks generated were found to be very significantly differently expressed(P<0.001) between the two groups.And 14 peaks were up-expressed while the other 10 peaks were down-expressed in COPD patients.The two max differentially expressed peaks were m/z:1012.92 and m/z:4152.85.Conclusion We were able to identify proteomic patterns that could clearly distinguish between COPD patients and healthy controls based on MB-WCX-MALDI-TOF-MS analysis.
