热疗对膀胱癌细胞BIU-87/HCPT耐药相关基因的影响

摘    要:目的观察热疗对耐羟基喜树碱膀胱癌细胞株BIU-87/HCPT多药耐药的逆转作用,并探讨其逆转肿瘤多药耐药的机制。方法不同温度条件下热疗、化疗、热疗联合化疗处理耐药膀胱癌细胞株BIU-87/HCPT后,用MTT法检测细胞增殖,流式细胞仪检测细胞周期、细胞凋亡、P-gp、多药耐药相关蛋白1(MRP1)、谷胱甘肽硫-转移酶π(GST-π)和survivin表达的变化。结果热疗对BIU-87/HCPT细胞具有抑制作用,呈温度依赖性且在43℃时最强。热疗可显著增加羟基喜树碱对BIU-87/HCPT细胞的抑制率,作用呈温度依赖性,热疗明显增加化疗的作用。热疗联合化疗明显减少P-gp、MRP1、GST-π和survivin的表达并呈温度依赖性。结论热疗可促进膀胱癌细胞株BIU-87/HCPT凋亡,增强化疗药物作用,部分逆转膀胱癌细胞的耐药性,减少P-gp、MRP1、GST-π和survivin的表达。

Abstract:Objective To investigate the effects and mechanism of hyperthermia in reversing multidrug resistance in BIU-87/HCPT cell line. Methods We treated BIU-87/HCPT cell by hyperthermia at different temperature(37 ℃-43 ℃),camptothecin and hyperthermia combined with camptothecin,and then detected the proliferation activity of BIU-87/HCPT cell by MTT assay.Expressions of P-gp,MRP1,GST-π and surviving as well as cell cycle and apoptosis were detected by flow cytometry. Results With the increase of temperature,the growth inhibition rate of BIU-87/HCPT cell rose significantly.Hyperthermia had an inhibitory effect on BIU-87/HCPT cell in the temperature-dependent manner with the strongest at 43 ℃.Hyperthermia could significantly enhance the inhibitory action of hydroxycamptothecin on BIU-87/HCPT cell.The percentage of apoptotic cells was significantly increased in hyperthermia group in the temperature-dependent manner.Hyperthermia significantly enhanced the effect of chemotherapy.The expressions of P-gp,MRP1,GST-π and survivin in BIU-87/HCPT cell were decreased significantly after hyperthermia combined with chemotherapy in the temperature-dependent manner. Conclusion Hyperthermia can enhance the inhibition rate of chemotherapeutic drugs,and partially reverse multidrug resistance and induce the apoptosis of BIU-87/HCPT cell with a decrease in expressions of intracellular P-gp,MRP1,GST-π and survivin.
