多囊卵巢综合征患者抗苗勒氏管激素与干细胞因子的相关性

摘    要:目的研究多囊卵巢综合征(PCOS)患者抗苗勒氏管激素(AMH)和干细胞因子(SCF)的表达情况及二者的相关性。方法选择年龄均小于35岁行体外受精与胚胎移植的PCOS患者102例,正常对照组90例,采用ELISA法检测血清和卵泡液中AMH和SCF的蛋白水平,分离并培养颗粒细胞;采用Real-time PCR检测PCOS患者颗粒细胞中AMH和SCF mRNA的相对含量,并对其进行相关性分析。结果①PCOS患者血清、卵泡液及颗粒细胞中AMH的相对表达较正常对照组明显升高,而SCF较对照组显著降低(P<0.01);②PCOS患者血清、卵泡液及颗粒细胞中AMH与SCF的表达均呈极为显著负相关(P<0.01)。结论多囊卵巢综合征患者血清、卵泡液及颗粒细胞中AMH与SCF的表达呈显著负相关。

Abstract:Objective To study the expressions of and correlation between anti-mullerian hormone(AMH) and stem cell factor(SCF) in patients with polycystic ovary syndrome(PCOS).Methods We recruited 102 patients with PCOS and 90 controls who all had received in vitro fertilization and embryo transfer(IVF-ET) at the age of less than 35 years.The levels of AMH and SCF protein in serum and follicular fluid were examined by ELISA;the relative mRNA expression in granulosa cells was detected by real-time PCR.The relationship between AMH and SCF expressions was analyzed by Pearson’s correlation.Results ① The expression of AMH was significantly increased in serum,follicular fluid and granulosa cells in PCOS group;however,SCF showed an obvious decrease in PCOS group compared with control group(P<0.01).② The expressions of AMH and SCF showed a significant negative correlation in serum,follicular fluid and granulosa cells of the patients with PCOS(P<0.01).Conclusion The expressions of AMH and SCF show a significantly negative correlation in serum,follicular fluid and granulosa cells of patients with PCOS.
